ObjectiveThis study aims to study and analyze the expression of autophagy related proteins(mitochondrial membrane junction receptor)FUNDC1 and BNIP3 in gastric cancer tissues.Firstly,the expression of FUNDC1 protein and BNIP3 protein in mitochondrial membrane junction receptor were preliminarily analyzed in gastric cancer tissues and para-cancer normal tissue samples paired with gastric cancer.Secondly,the positive expression of these two proteins in gastric cancer tissues was analyzed with clinical-related pathological indicators,such as age,tumor size,lymph node metastasis,TNM stage,etc,and the correlation of FUNDC1 and BNIP3 protein with the above indexes was discussed.Finally,through this experimental research,it is expected to find markers for the diagnosis of gastric cancer and provide more basic clinical experimental theoretical basis for the treatment of new targeted molecular drugs for gastric cancer.Methods From January 2017 to December 2018,80 surgical specimens of gastric cancer patients in the Lanzhou University Second Hospital were collected as the experimental group.All the included tissue specimens had complete clinicopathological data and follow-up data,ranging from 30 to 90 years old,with an average age of 68 years old.All selected tissue samples were fixed with conventional fixative solution(formalin),embedded,sliced,removed and prepared for immunohistochemical labeling(IHC).The proportion of staining range and intensity of tumor cells in tissue sections are the main basis for IHC final interpretation.Two experienced chief physicians(deputy)of pathology department in the Lanzhou University Second Hospital read the film for grading,and adopted the semi-quantitative grading method for the expression of BNIP3 and FUNDC1 protein.Statistical treatment:usec~2test to analyze whether there is a difference in the immunohistochemical expression of BNIP3 and FUNDC1 protein in gastric cancer between cancer and adjacent tissues.Spearman rank correlation was used to analyze the correlation between BNIP3 and FUNDC1,and Spearman rank correlation and Pearson rank correlation were used to analyze the correlation between BNIP3,FUNDC1 and the pathological characteristics of gastric cancer patients respectively.All the data were statistically analyzed and proofread for three times,and then SPSS16.0 was recorded for statistical analysis.The data were reviewed by the epidemiologist,and the images were edited and processed by Graphpad prism 6.Results1.BNIP3 protein and FUNDC1 protein were lightly browned in the cytoplasm of normal tissues adjacent to gastric cancer,showing low expression,while no obvious expression was found in the nucleus or cell membrane.Mitochondrial membrane junction protein BNIP3 protein and FUNDC1 protein were mainly displayed in the cytoplasm of cells in the gastric cancer tissue samples,showing dark brown color and high expression.2.The expression of mitochondrial membrane junction protein BNIP3 and FUNDC1 in cancer tissue samples and normal tissue samples was stained by immunohistochemical monoclonal antibody:among the 80 cases of gastric cancer tissues in the Lanzhou University Second Hospital,53 cases showed high expression of BNIP3,with a high expression rate of 6.25%.Among the 80 paired normal tissues adjacent to gastric cancer,36 patients showed high expression of BNIP3,with a high expression rate of 45.0%.In the gastric cancer tissue samples,the high expression rate of mitochondrial membrane connector protein BNIP3 was significantly higher than that of its paired paracancerous normal tissues,and the difference was statistically significant at the test level of 0.05(c~2=5.71,P=0.016).In the gastric cancer tissues with the same number of cases,the mitochondrial membrane junction protein FUNDC1 was highly expressed in 59 cases,with a high expression rate of 73.7%.In the paired normal tissue samples,only 38 cases showed high expression of FUNDC1,with a high expression rate of 47.5%.The expression level of FUNDC1 protein in the IHC colored mitochondrial membrane junction in the cancer tissue sample was significantly higher than that in the paired para-cancer normal tissue sample,and the difference was statistically significant at the test level of=0.05(c~2=11.54,P=0.018).3.In the 80 gastric cancer samples,40 cases were positive for BNIP3 and FUNDC1 protein,17 cases were negative for BNIP3 and FUNDC1 protein,two protein expression was moderately correlated and positively correlated(r=0.664,P<0.001).4.In gastric cancer specimens,the high expression of mitochondrial membrane junction related protein BNIP3 was positively correlated with the metastasis of gastric cancer parenchymal cells through lymph nodes(r=0.226,P=0.013),which was statistically significant.The high expression of FUNDC1,a protein related to mitochondrial membrane junction,was also positively correlated with lymph node metastasis(r=0.308,P=0.006)was statistically significant.Conclusions 1.Mitochondrial membrane junction protein-BNIP3 protein and FUNDC1 protein were both highly expressed in gastric cancer tissues.2.The high expression of BNIP3 protein and FUNDC1 protein in gastric cancer tissues is closely related to the metastasis of gastric cancer cells through lymph nodes.3.There was a positive correlation between the expression of BNIP3,FUNDC1positive protein in gastric cancer tissues.
