As a subtype of breast cancer,triple-negative breast cancer has a high incidence in young women and has a high recurrence rate and metastasis.Cancer is a complex disease caused by the high expression of specific genes.One of the main reasons why triple-negative breast cancer is difficult to cure is because of the large amount of Survivin inhibitory protein in its tumor cells,which greatly inhibits the programmed cell death of tumor cells after stimulation by chemotherapy or other therapeutic means.Thus,triple-negative breast cancer produces therapeutic resistance.Therefore,Survivin can be an ideal target gene for tumor gene therapy.In recent years,with the rapid development of molecular biology technologies,people have conducted in-depth research on the pathogenesis of breast cancer,and gene therapy has gradually become a therapeutic model with good application prospects.In this paper,we used a tumor gene antagonism strategy to down-regulate the expression of Survivin gene in triplenegative breast cancer cells by deoxyribozyme technology using a highly efficient,safe and specific Survivin deoxyribozyme(DNAzyme)to reduce the resistance of tumor cells,apoptosis ability,while inhibiting further invasion and metastasis of tumor cells.For the effective delivery of chemotherapeutic drugs,we chose to coat a layer of mesoporous silica on the outside of the nanogold rod,load the chemotherapeutic drug DOX,and use the unique photothermal conversion characteristics of the gold rod to impart photothermal properties to the material.In addition,the thermophilic small molecule(ACVA)coupled to the deoxyribozyme is used to improve the stability of the mesoporous silicon surface and achieve fixed-point release under near-infrared light.Through a series of measures,the expression of Survivin gene in triplenegative breast cancer can be reduced,the probability of tumor cell apoptosis can be increased,the tolerance of tumor cells can be reduced,and the metastasis ability of tumor cells can be inhibited.The effect of triple-negative breast cancer treatment has been significantly improved.
