Objective: Preliminary effect of 125-iodine particle radiotherapy on animal model of echinococcosisMethods:(1)A total of 20 athymic nude mouse;weighting about 20g;10 weeks old;taken from the subcutaneous part of the right neck of nude mouse,the needle of the syringe was obliquely inserted into the subcutaneous lesion model of nude mouse.The nude mouse that were successfully modeled were randomly divided into experimental group(n=10),control group(n=5),and blank control group(n=5).In the experimental group,nude mice were implanted with radioactive 125-iodine particles,and the size of the lesions before implantation was measured.The control group received a puncture needle and the lesions were not implanted;the blank control group did not interfere with the intervention.Conventional breeding after surgery.The size of the lesions in nude mouse was measured daily,and the model was sacrificed 45 days later.The lesions were weighed,the tissue was examined by trypan blue staining and the pathological results were obtained by HE staining.(2)A total of 14 rats,weighing about 600 g,12 weeks old.After anesthesia and laparotomy,the multi-atrial echinococcosis suspension was implanted in the liver,and the rat model of multi-room echinococcosis was established.14 successful model rats were divided into experimental group and control group.After 45 days of experimental intervention,the rats were sacrificed.Observing the activity of the protoscolex and the red-hematoxylin(HE)staining were used to observe the pathological changes under the microscope.Results:1.Nude mouse model(1)After 6 months of inoculation,the success rate of modeling was 80%(16/20).After 45 days of experimental intervention,8 model(8/10)survived in the experimental group,and 4 model in the control group survived(4/ 5)The blank control group survived 4 mice(4/5).(2)After 45 days of particle experiment intervention,the diameters of the nude mice in the experimental group and the control group were 0.84 ± 0.45 cm and 3.14 ± 0.39 cm,respectively(t = 6.477,P < 0.001).The diameters of the nude mice in the experimental group and the blank control group were 0.84 ± 0.45 cm and 2.66 ± 0.88 cm,respectively(t = 4.355,P < 0.002).(3)After the intervention of the experimental group and the control group,the activity and quantity of the original head of the experimental group were significantly reduced compared with the control group and the blank control group.(4)In the experimental group,the number of Echinococcosis showed significant death and decreased under HE staining.2.Rat model(1)After 6 months of inoculation,the success rate of modeling was 100%(14/14).Radioactive particles were implanted in the orthotopic lesions of the liver after anesthesia,and 10 rats(10/10)survived in the experimental group.The rats survived 4(4/4).(2)After the execution of the lesion tissue,the protoscolex section was extracted,and observing the activity of the protoscolex under the microscope;(4)The experimental group and the control group were treated with liver lesions and the pathological changes under the microscope were observed.Conclusion: The experimental results suggest that 125-iodine particles can kill protoscolex and inhibit the further development of vesicular hydatid disease through radiation effects.
