Objective: This thesis aims at investigating the expression of angiogenesis factor mRNA in chronic myeloid leukemia K562 cell under hypoxia condition and exploring the relationship between the concentration of vascular factors and HIF-1? in cultured supernatant in vitro.Moreover,the regulating effect of hypoxia on vascular factor formation in chronic myeloid leukemia by hif-1? will be discussed,thus enriching the basic theory of angiogenesis regulation of chronic myeloid leukemia under hypoxic conditions.Methods: Selected as the experimental object,the K562 cells were transfected with the Lentivirus virus liquid with hif-1? interference.Then the transfection was observed by inverted immunofluorescence microscope,and the K562 cells were treated with hypoxic culture as hypoxic interference group after transfection.In order to compare,the K562 cells without virus were cultured by normal oxygen and hypoxia as the regular oxygen group and the hypoxic group respectively.Through the above treatment,the cells and culture supernatant were collected,and the RNA was extracted to detect HIF-1?,VEGF,Ang-I,Ang-II,TGF-? and TGF-? mRNA expression by RT-PCR.Furthermore,the concentrations of VEGF,Ang-I,Ang-II,TGF-? and TGF-? in the cultured supernatant were detected by ELISA.Results: The levels of VEGF,Ang-II and TGF-? mRNA in the hypoxic group were higher than that in the regular oxygen group,while the TGF-? mRNA in the hypoxic group was lower than that in the normal oxygen group.The TGF-? was not detected in the culture,and the concentration of VEGF and Ang-II in the low oxygen group of was higher than that of the oxygen group,while the TGF-? concentration was higher than that in the hypoxic group(p<0.05).The expression of VEGF,Ang-II and TGF-? mRNA in hypoxic group was higher than that in hypoxic disturbance group,while the expression of hypoxic disturbance in TGF-? mRNA was significantly higher than that of hypoxic group,and the TGF-? was not detected in the supernatant.The concentration of VEGF,Ang-II,and TGF-? in the hypoxic group was higher than that of hypoxic disturbance group(p<0.05).Conclusions: 1.HIF-1? mRNA positively regulates the expression of VEGF,Ang-II and TGF-? mRNA in K562 cells and promotes the ability of VEGF,Ang-II self-secretion;2.HIF-1? mRNA inhibits the expression of TGF-? mRNA,but promotes its autocrine function;3.The effects of hypoxia by HIF-1? on the expression of the angiogenesis factor and autocrine of mRNA in K562 cells were not identical.
