Objective: To observe the effect of electric acupuncture method on the muscle satellite cell and the signal transduction pathway of TLR4/MyD88/NF-?B in cervical spondylosis model rats.Methods: Tirty-two Wistar rats were used in this study,all of them are male.Nine rats were randomly selected as blank control group(referred to as blank group).Except the blank group,all the other rats carried on the cervical spondylosis model.Placed the rats in special cervical anteflexion fixed frame,with the head and neck are fixed in forward bends a 45°Angle.Ensure that the degree of fixation tightness does not affect the normal breathing of rats.The rats were fixed for 5 hours once a day for 3 months.At the end of modeling,two rats were randomly selected from the blank group and the rats after modeling.Transmission electron microscopy was used to detect the morphological changes of the posterior cervical muscle.After the modeling was completed,the rats were randomly divided into 3 groups,7 rats in each group.They were: model control group(referred to as model group),model electroacupuncture group(referred to as electroacupuncture group),model meroxicam group(abbreviated as meroxicam group).Electroacupuncture group: Adopt self-made elastic band method for safety fixation.According to "experimental acupuncture and moxibustion" edited by li zhongren and comparing anatomical methods,"fengchi acupoints"(bilateral)were selected.The two "fengchi acupoints" were connected with the stimulation instrument of han's meridians,and adjusted the stimulation parameters with Density wave,voltage 2-4V,current 2mA,and frequency 2/100 hz.Treatment time: 25 min,once a day.A course of 10 days,a total of 2 courses of treatment.Meroxicam group: intragastric intervention,the amount of mobectin tablet is 0.7878 mg/kg a day,as the same time with the electroacupuncture group;model group: no treatment,remaining with the electroacupuncture group.After the intervention course,the morphological changes of the posterior cervical muscle were observed under electron microscope.The contents of PCNA,MyoD,TLR4,MyD88 and NF-?Bp65 were determined by immunohistochemistry.To investigate the effect of electric acupuncture method on the muscle satellite cell and the signal transduction pathway of TLR4/MyD88/NF-?B in cervical spondylosis model rats.It is suggested that electroacupuncture can promote the proliferation of myosatellite cells and may be achieved by mediating the anti-inflammatory effect of TLR4/MyD88/ NF-?B signaling pathway.Results:(1)Electron microscopy: blank group: The myofilaments were punctate in cross-section,the mitochondria were normal,and the sarcomere and myoserous membrane were intact.Myofibrils were closely arranged,with obvious transverse stria,A,I,Z,H,and M lines in regular alternating arrangement.The serous membrane was intact without rupture,the nuclear type was normal,the nuclear membrane was smooth,and the mitochondria were evenly distributed.Model group: myofibril structure discontinuous and chaotic,individual mitochondria swelling.A small amount of fiber and sarcomere fracture,with muscle space close to normal.Electroacupuncture group: myofibrils were clearly visible,and myofilaments were punctate in cross-section.Sarcomere and serosa were intact,individual mitochondria were swollen,and a few of them had vacuoles at the break of myofibrils.Meroxicam group: the structure of myofibrils was discontinuous and disordered.Transverse stria is apparent,individual mitochondria are swollen,among them a few are with the break place of flesh fibril vacuole.(2)Immunohistochemistry(IHC): Compared with the blank group,the positive expression levels of PCNA and MyoD in the model group were lower,and the positive expression levels of TLR4,MyD88 and NF-?Bp65 were significantly increased,and the differences were statistically significant(P<0.05).Compared with the model group,the protein expressions of PCNA and MyoD were significantly increased in the electroacupuncture group and the meroxicam group,while the protein expressions of TLR4,MyD88 and NF-?Bp65 were significantly decreased,and the differences were statistically significant(P<0.05).Conclusion:(1)Electroacupuncture at "fengchi acupoints" could up-regulate the expression of muscle satellite cells.(2)The expression of TLR4,MyD88 and NF-?Bp65 in muscle cells could be down-regulated by electroacupuncture at "fengchi acupoints".(3)Promote the proliferation of muscle satellite cells and inhibit the inflammatory response of muscle cells by electroacupuncture therapy.This mechanism may be down-regulated the positive expression ofTLR4,MyD88 and NF-?Bp65,probably due to the regulation of the TLR4/MyD88/ NF-?B signal transduction pathway.
