This dissertation is composed of three chapters.The chemical constituents and their bioactivities from three fungal species Antrodiella albocinnamomea,Bipolaris eleusines,and Ganoderma orbiforme were investigated.By using normal phase silica gel column,RP-18,gel permeation chromatography,MPLC,HPLC,etal.chromatographic methods,and extensive spectroscopic analysis(including NMR,MS,UV,IR,X-ray diffraction),a total of85 compounds,including 28 new compounds,were obtained,which were ascribed into sesquiterpenes,triterpenes,benzothiazole,peptides,etc.Chapter 1 describes chemical and biological studies on higher fungus Antrodiella albocinnamomea.A.albocinnamomea is a wood-roting fungus belongs to Polyporaceae family.A total of 29 compounds were isolated from the broth of A.albocinnamomea,including 13 new compounds which involved in triquinanes and illudane type sesquiterpenes.Antroalbocin A,a new carbon skeleton compound which has a bridged tricyclic system.Biogenetically,antroalbocin A should be derived from triquinane-type sesquiterpenoid by a key step of an alkyl migration.Antroalbocin A showed certain antibacterial activity to Staphylococcus aureus,which makes it a good scaff old for further investigations.Chapter 2 deals with the chemical investigations on potato endophytic fungus Bipolaris eleusines.In this study,a chemical investigation on the culture of B.eleusines led to the isolation of 33 compounds including 8 new ones.Sativenes and seo-sativenes sesquiterpenes are main classes of the isolates.Of them,bipolathizole A(38)was promised as a new carbon skeleton which is noteworth for the C-C linkage of a sativene sesquiterpene and a phenylcyclo thiazole ring.Some selected compounds were evaluated for their antibacterial activities on four potato pathogenic fungi Phytophthora infestane,Alternaria solani,Rhizoctonia solani and Fusarium oxysporum.As a result,bipolenin D? F(33? 35)and bipolenin H(37)showed inhibitory activities against Fusarium oxysporum with minimum inhibitory concertrations(MIC)of 25,50,100,and 12.5 ?g/mL,respectively.Chapter 3 elaborates chemical investigations on Ganoderma orbiforme.In the current study,a total of 23 compounds including 5 new compounds were isolated from the fruiting bodies of G.orbiforme.Ganoboninone G(65)and ganomycin I(85)showed weak inhibitory activities against M.tuberculosis H37 Ra.Ganomyin I(85)also showed inhibitory activity against Gram-positive bacteria,B.cereus,E.faecium,and S.aureus,with MIC values of 25 ?g/mL,25 ?g/mL,and 12.5?g/mL,respectively.As indicated by the result,lanostane triterpenoids were considered as the main secondary metabolites of G.orbiforme.
