Rheumatoid arthritis?RA?is a multi-systemic autoimmune disease characterized by chronic inflammation of the joints,synovial inflammation,joint destruction and so on.Its main characteristics are synovial inflammation,cartilage and bone progressive destruction.Clinical manifestations of bone and joint deformity,ankylosis,loss of function,to human health and quality of life has caused a serious impact.The production of pro-inflammatory factors,in inflammatory synovial tissue of rheumatoid arthritis can affect the activity and differentiation of osteoclasts.The Sargentodoxae Caulis has the function of clearing heat and detoxification,promoting blood circulation,dispelling wind and relieving pain.For hot ulcer,intestinal abscess abdominal pain,amenorrhea,dysmenorrhea,tumbling swelling pain,rheumatic arthralgia.In this study,we investigated the effects of the total phenolics of Sargentodoxae Caulis on the bone destruction of rheumatoid arthritis,in order to provide a theoretical basis for its application in the prevention and treatment of RA.The monomers compound was isolated from the ethyl acetate layer of the Sargentodoxae Caulis and identified as?-?-epicatechin by modern spectroscopic techniques such as ESI-MS,1H-NMR,13C-NMR,DEPT,ROSY.To establish a method for the determination of the content of succulent?-?-epicatechin,the total phenol was enriched and purified.Through the static adsorption and analytical experiment,the adsorption rate and the resolution rate were taken as the indexes,respectively,from the HZ818,AB-8,SP825,HP20,XAD16N Among the five macroporous adsorption resins the optimum macroporous adsorption resin was selected for enrichment and purification of SP825,and the total phenol of Sargentodoxae Caulis content was determined to be 67.31%by ultraviolet-visible spectrophotometry;The content of?-?-epicatechin in total phenol of the Sargentodoxae Caulis was determined by high performance liquid chromatography?HPLC?,and the content of?-?-epicatechin was 3.48%.Osteoclasts were obtained by primary isolation of rabbit bone marrow mesenchymal cells by 1?,25-?OH?2VitD3 induction.The number of TRAP positive cells,the activity of TRAP enzyme and the area of bone lacuna were used as indexes.To study the effect of total phenol of Sargentodoxae Caulis on osteoclast differentiation and bone resorption activity.The results showed that the total phenol of Sargentodoxae Caulis were inhibited differentiation of osteoclast and the activity of bone resorption,and the inhibition was most significant at the dose of 31.25?g/ml.Ovariectomy combined with bovine type II collagen induction,With the rat body weight,toe swelling,arthritis index score,spleen coefficient and serum OCN,E2,TRAP,IL-1,IL-6,TNF-?,IL-17,Ca,P and ALP as the detection index,combined with joint imaging structure and joint histopathological analysis,establish a rat kidney deficiency CIA model.The results showed that compared with the normal group,the weight of kidney deficiency CIA model rats decreased,joint swelling and arthritis score index increased,spleen index decreased,and serum the levels of OCN,E2 and Ca concentration decreased,The concentration of TRAP,IL-1,IL-6,TNF-?,IL-17,P and ALP increased,and the degree of joint pathological damage and bone destruction increased significantly.Using the above model,With the rat body weight,toe swelling,arthritis index score,spleen coefficient,serum E2,TRAP,IL-1,IL-6,TNF-?,IL-17,Ca,P,ALP,etc.for detection the index,combined with joint imaging structure and joint histopathology analysis,studied the effects of total phenols of Sargentodoxae Caulis on the serum factors,joint histopathology and bone destruction of kidney deficiency CIA rats.The results showed that the total dose of total phenols of Sargentodoxae Caulis is significantly reduced the toe swelling,arthritis index score and rat spleen coefficient of kidney deficiency CIA rats.and the high dose group and the middle dose group had obvious effects;High and medium dose groups of total phenol of Sargentodoxae Caulis could increase the levels of serum E2 and Ca,and inhibit the TRAP,IL-1,IL-6,TNF-?,IL-17,P and ALP concentrations.reduced bone destruction and pathological damage in kidney-deficiency CIA rats.In summary,the high and medium does of total phenol of Sargentodoxae Caulis could inhibit osteoclast activity and bone resorption,and has a certain alleviation effect on bone destruction of rheumatoid arthritis.
