Synthesis And Antitumor Activity Of 2,5-disubstituted 1,3,4-oxadiazole Derivatives

Among heterocyclic compounds,1,3,4-oxadiazole has become an important structural motif for the development of new drugs.Compounds containing 1,3,4-oxadiazole nuclei have a wide spectrum of biological activities,including antimicrobial,antifungal,anti-tuberculosis,analgesic,anti-inflammatory,anti-virus,anti-cancer,anti-hypertension,anti-convulsion,anti-diabetes,insecticidal and plant growth regulation.1,3,4-oxadiazole derivatives have unique structure and great biological potential.In this paper,2,5-diphenyl-1,3,4-oxadiazole derivatives were synthesized by expanding the structure of 1,3,4-oxadiazole molecule,hoping to obtain new compounds with better biological activity and less toxic side effects.Using substituted benzoic acid,p-bromobenzoic acid,p-methyl benzoyl hydrazide,p-bromobenzoyl hydrazide and substituted benzoic acid as starting materials,10 derivatives containing 1,3,4-oxadiazole ring were synthesized by Suzuki coupling reaction,cyclization reaction and reduction reaction.The structures of all compounds were characterized by 1H NMR,13 C NMR and high resolution mass spectrometry.MTT colorimetric assay was used to determine the inhibitory activities of2,5-disubstituted-1,3,4-oxadiazole derivatives on the growth of human lung cancer cell lines(A549),human hepatoblastoma(HepG2),experimental proliferating epidermal carcinoma cell lines(HeLa)and human breast cancer cell lines(MCF-7).The results showed that the target compounds had different inhibitory activities on four kinds of cancer cells.However,some compounds have a negative inhibitory effect on four kinds of cancer cells,that is,they can promote the proliferation of cancer cells.
