| Objective: To investigate the effect of neural stem cell transplantation on the level of inflammation in rats with cerebral ischemia-reperfusion injury and the effect of neural stem cells transplantation on functional recovery of injured nerve in rats.Methods: Ischemia-reperfusion model of rats was built by middle cerebral artery occlusion and reperfusion(MCAO/R,occluded 90min)method.Rats were randomly divided into sham operation group(SHAM group),phosphate buffer(PBS)control group,and NSCs transplantation group.Then at the 1st,3rd,14 th and 28 th days after stroke and transplantation,the neurological functions were assessed by m NSS score,rotarod test,infarct volume,the cerebrospinal fluid were collected and then detected by ELISA.Results:(1)The neurological function of NSCs transplantation group was significantly improved than PBS group,and the difference had statistical significant(P<0.05).(2)The infarct volume of NSCs transplantation group was significantly smaller than PBS group,and the difference had statistical significant(P<0.05).(3)The levels of TNF-αand IL-1βin cerebrospinal fluid of NSCs transplantation group were lower than those in control group at 1st,3rd and 14 th days,and the levels of TGF-β1 and IL-10 in NSCs group were higher than that in control group.The difference had statistical significant(P<0.05).Conclusion: Transplantation of NSCs can alleviate the inflammatory response and reduce the damage of nerve cells after cerebral ischemia reperfusion injury and promote the recovery of nerve function after cerebral ischemia-reperfusion in rats. |
