Objective: To investigate the effects of electroacupuncture(EA)and pregabalin on cognition impairment induced by trigeminal neuralgia(TN)in rats.Methods: Rats were randomly divided into 4 groups: sham group,cobra venom group,EA group and pregabalin group.The TN model was induced by the administration of cobra venom to the left infraorbital nerve.Free behavioral activities was observed 3 days before the operation and 3,6,9,12,15,20,30,40 days after the operation.On postoperative day 14,EA and pregabalin was administered in two groups respectively.The abilities of spatial learning and memory were determined in the Morris water maze.The ultrastructural alterations of the gasserian ganglion,medulla oblongata and hippocampus were examined via electron microscopy.The changes on long-term potentiation(LTP)was investigated.Results:(1)After treatment,exploratory behavior increased and grooming behavior decreased(P<0.05)compared with cobra venom group.(2)Moreover the demyelination of neurons in gasserian ganglion and medulla oblongata were revised while the number of myelinating fibers showed an increase.(3)The number of platform site crossing and the average percentages of the time in the target quadrant increased in EA group compared to cobra venom group,however,pregabalin group showed no difference with cobra venom group.(4)The field excitatory postsynaptic potential(fEPSP)slopes increased(P<0.05)in EA group compared to cobra venom group and the field excitatory postsynaptic potential(fEPSP)in pregabalin group slopes showed no difference with cobra venom group(P>0.05).(5)Vacuolation degeneration in hippocampus neurons in EA group was mild while it was severe in pregabalin group.Conclusion:(1)In this research,we found that TN could result in cognition dysfunction.(2)EA and pregabalin could alleviate the TN induced by cobra venom.(3)However,EA can also inhibited the cognition deficit induced by TN while pregabalin can not.
