Paeoniae Radix Alba,the roots of Paeonia lactiflora Pall.,is a common traditional Chinese medicine(TCM)that is used for nourishing blood and regulating menstruation,astringing Yin to check sweating,emolliating the liver,relieving pain,and pacifying and repressing the liver Yan.In this study,the suitable drying processing methods,optimization of extraction technology using response surface methodology and application of HPLC fingerprint combined with QAMS in quality evaluation of Paeoniae Radix Alba were analyzed and discussed.The main contents and conclusions were as follows:1.Simultaneously determine 4 monoterpene glycosides and 2 phenolic acids in Paeoniae Radix Alba with different drying methods(sun drying,shade drying,hot air drying,microwave drying,far infrared drying and vacuum freeze drying)by HPLC.PCA was carried out by SPSS software to evaluate the quality of different processed products from Paeoniae Radix Alba.The results showed that the contents of chemical components were in accordance with the requirements of Pharmacopeia of the People's Republic of China(The First Division of 2015 Edition).The highest total contents of monoterpene glycosides and phenolic acids in Paeoniae Radix Alba processed by vacuum freeze drying were 6.2% and followed by processed by drying with hot air at 60 ?(6.1%).According to the PCA results,the comprehensive scores of monoterpene glycoside and phenolic acid compounds in Paeoniae Radix Alba processed with different drying methods in the order from high to low were vacuum freeze dried > dried with hot air at 60 ? > microwave dried > far infrared dried > sun dried > dried with hot air at 40 ? > dried with hot air at 80 ? > shade dried > dried with hot air at 100 ?.In conclusion,drying with hot air at 60 ? was the appropriate drying method of Paeoniae Radix Alba by comprehensive analysis of the content,cost and practicality.2.Application of Box-Behnken experimental design and high performance liquid chromatography fingerprint to optimize the extraction process of Radix Paeoniae Paeoniae.The total common peaks area of HPLC fingerprints were regard as a evaluation index for the selection of optimal conditions by using response surface methodology.The optimum ultrasonic extraction conditions were as follow: the ultrasonic extraction time was 50 min,the ethanol concentration was 67.31%,the solvent-solid ratio was 17.17 mL?g and the ultrasonic extraction time was 50 min.The optimized conditions were validated and close agreement was observed with the validation experiment and predicted value.It concluded that HPLC fingerprints combined with the Box-Behnken experimental design could be used to optimize extracting process of Paeoniae Radix Alba.3.Paeoniflorin was selected as markers of ingredients to establish HPLC fingerprint of Paeoniae Radix Alba,and the relative correction factors of peoniflorin to gallic acid,catechin,albiflorin and benzoylpaeoniflorin were calculated by slope correction method.Simultaneously,the five components were determined by external standard method.The accuracy and feasibility of QAMS were evaluated by comparison of the results between external standard method and QAMS.Ten common peaks were identified in the HPLC characteristic fingerprint,and five components were verified in twenty-eighth batches of Paeoniae Radix Alba.Compared with the results obtained using the ESM,QAMS method was found to have no significant differences as verified by the T-test(P > 0.05,n = 28).The methods of HPLC fingerprint combined with QAMS can be apply to assess the quality of Radix Paeoniae Alba materials.
