Study On The Functional Mechanism Of Linc00857 In Lung Adenocarcinoma

Lung cancer is one of the most deadly cancers in humans and poses a great threat to the human health.Diagnosis in the early age is important for the treatment of lung cancer,but there is still a lack of effective and broad-spectrum biomarkers that can be used for the early diagnosis.Therefore,the exploration of novel biomarkers for diagnosis about lung cancer and further to study the mechanisms underlying the drive mutations in lung cancer are need.Recently,many lncRNAs that are differentially expressed between normal tissues and cancer tissues have been found to play an important role in the development of cancer,which have attracted increasing attention.Among them,linc00857 has been found to be highly expressed in lung adenocarcinoma,which could promote the cell proliferation and migration.In the present study,the datas about lung cancer from TCGA database were used to analyze the lncRNAs that were significantly differently expressed between normal tissues and lung adenocarcinoma tissues.It was found that linc00857 was abnormally overexpressed in lung adenocarcinoma.So,next we focused on linc00857 and to characterize it.Firstly,the exact length of linc00857 was cloned by RACE technology.It was found that it had no coding capacity but had a high transcription activity.Besides,it was less conservative.After nucleocytoplasmic isolation of lung adenocarcinoma cells,linc00857 was quantified and found to be concentrated in the cytoplasm.Based on the the results of cell proliferation experiments and so on,it was confirmed that it promotes the cell proliferation of lung adenocarcinoma cells.In order to find the molecular mechanism of linc00857 in lung adenocarcinoma,we constructed the ceRNA network of linc00857 and verified it by experiments.The results show that linc00857 competitively ARHGAP5 by binding to miR486-5p,thereby increasing the expression of p190 B RhoGAP and promoting cell proliferation and metastasis.Additionally,we analyzed the exosomes secreted by lung adenocarcinoma cells and their biological function,we found that linc00857 can be embedded into exosomes and came into the recipient cells through exosomes,thereby promoting cell proliferation.This indicates that the clinical condition of lung adenocarcinoma patients may be judged by detecting the content of linc00857 in exosomes.
