Objective The changes of Zn2+ concentration and the expression of zinc transporter Zip10 and ERS factors in HFp EF rats were discussed.Methods Based on the rat model of HFp EF by dietary zinc intervention,detection of Zn2+concentration in serum and myocardial tissue of rats in the experimental and control groups: Another member of our group has used the DSS rat to construct an animal model of HFp EF in the previous experiment.DSS rats were randomly divided into 5groups(12 in each group),model/experimental group [4 groups,including high-salt low-zinc diet group(HL),high-salt normal zinc diet group(HN),high-salt high-zinc diet group(HH90),and high-salt pole high-zinc diet group(HH270)] and control group [normal salt normal zinc diet group(NN)],in which the sodium salt content of the high-salt diet in the model group was 8%,and the zinc content in each group was10 mg/kg,30 mg /kg,90mg/kg and 270mg/kg,the normal salt of the normal salt in the control group,the content of sodium chloride is 0.3%,the content of zinc is30mg/kg,the content of the remaining feed components and the conditions of feeding were the same between the each groups.Then,from 7 weeks old to 20 weeks old,the model was evaluated by echocardiography,wet lung/dry lung ratio,serum and myocardial tissue BNP content.Rats in the experimental group who met the diagnostic criteria of the HFp EF model and the control group were enrolled and the heart and serum samples were taken.Determination of Zn2+ concentration in serum and myocardial tissue of each rat by ICPOES.The expression of Zip10 in the myocardial tissue of the test and control rats:q PCR and WB were used to detect the Zip10 m RNA and protein expression in rat myocardial tissue.The detection of ERS in the myocardial tissue of the test and control rats:q PCR and WB were used to detect the m RNA and protein expression of ERS factors such as CHOP and Bip in rat myocardial tissue.The expression of Zip7 in the myocardial tissue of the test and control rats:q PCR and WB were used to detect the Zip7 m RNA and protein expression in rat myocardial tissue.Results The results of ICPOES showed that the concentration of Zn2+ in the myocardial tissue of the experimental group was significantly higher than that of the control group(P<0.05).There was no significant difference between the experimental groups(P>0.05).The serum Zn2+ concentration in the experimental group was significantly lower than that in the control group,and the difference was statistically significant(P<0.05).The results of q PCR and WB showed: that the m RNA and protein expression levels of Zip10 in the myocardial tissue of the experimental group were significantly lower than those in the control group(P<0.05).There was no significant difference between the experimental groups.(P>0.05).that the m RNA and protein expression levels of ERS factors such as CHOP and Bip in the experimental group were significantly higher than those in the control group(P<0.05).There was no significant difference between the two groups(P>0.05).The m RNA and protein expression levels of Zip7 in the experimental group were slightly higher than those in the control group,and the difference was not statistically significant(P>0.05).Conclusion1.The concentration of Zn2+ in rat myocardial tissue increased and reduced in serum in HFp EF rats.2.The m RNA and protein expression of zinc transporter Zip10 was down-regulated in rat myocardial tissue in HFp EF rats.3.Endoplasmic reticulum stress levels were elevated in rat myocardial tissue in HFp EF rats.4.Dietary zinc intervention had no effect on Zinc ion concentration in myocardial tissue,transporter Zip7?Zip10 expression and ERS level in HFp EF rats.
