Vascular aging begins in early life and is a series of degenerative changes that occur with ageing vascular structure and function.The degree of vascular aging is a key factor in determining the health status of the elderly and is very important in aging research.Therefore,research on delaying vascular aging is of great significance.Studies have found that high glucose can cause vascular cell senescence and aging of blood vessels.In previous studies,it was proved that the extract of Ginseng,Sanqi and Chuanxiong can delay vascular aging.Therefore,this experiment established a mouse aortic aging model induced by high glucose.The mouse aorta was studied for the study,in order to observe the effect of high glucose on the aorta of mice and the changes in the process of drug intervention,to clarify the effect of high glucose on aortic aging in mice and the mechanism of extract of Ginseng,Sanqi and Chuanxiong on aortic aging.Objective: In this study,the mouse aorta was used as the research object,and the protective effect of the extract of Ginseng,Sanqi and Chuanxiong on vascular aging induced by high glucose in mice was studiedfrom AMPK/mTOR pathway.Method: A total of 130 male C57BL/6 mice were randomly divided into a blank control group and a high glucose group.The high glucose group was intraperitoneally injected with STZ and fed continuously for seven months with a high-fat diet.The mice were divided into 4 groups:model group,traditional Chinese medicine low-dose(0.819g/kg),high-dose groups(1.638g/kg)and the metformin group(150mg/kg),the mice were intragastrically administered once a day for nine weeks,and the changes in body weight and blood glucose were measured.In the open field test,the mice were explored for spontaneously of exercise activity,and the high-resolution small animal ultrasound imaging system was used to observe the carotid morphology of the mice.At the end of the administration,HE staining and Masson staining were used to detect the morphology of the thoracic aorta in mice.The AGE and MMP-2 levels were detected by immunohistochemistry to identify the degree of aortic aging in mice,and the expression of related inflammatory factors was detected by Bio-plex suspension chip system.Western blot was used to detect the expression of p16,p21,AMPK,p-AMPK,mTOR,p-mTOR,LKB1,p-LKB1,p70s6 k and p-p70s6 k proteins in mouse thoracic aorta.Result: Compared with the blank control group,the mice in the model group were dull and slow-moving and withered;the weight were loss and the blood glucose were elevated(P<0.01);the total motor distance andcentral activity time decreased(P<0.01);the carotid artery wall thickness were increased(P<0.01),and the blood flow velocity were decreased(P<0.01).HE and Masson staining showed that the intimal injury was severe and hyperplasia,the middle membrane was obviously proliferated and the arrangement was not neat,the elastic fiber was disordered and broken,and the collagen content increased(P<0.01);The brown deposition of AGE and MMP-2 in the aorta increased(P<0.01);serum inflammatory factors IL-1?,IL-6,TNF-? increased(P<0.01);the expression of p16,p21,mTOR,p-mTOR,p70s6 k,p-p70s6 k protein was increased(P<0.01),and the expression of AMPK,p-AMPK,LKB1 and p-LKB1 protein was decreased(P<0.01).Compared with the model group,there was no significant difference in body weight between the groups,and the blood glucose level decreased significantly(P<0.05);the total exercise distance and central activity time increased(P<0.05);the carotid wall thickness decreased(P<0.05);blood flow velocity increased(P <0.05);HE and Masson staining showed that the intimal injury was not obvious in each group,the medium membrane was less proliferated,the collagen fiber content was decreased(P<0.05);the expression of AGE and MMP-2 was decreased(P<0.01);the serum inflammatory factor IL-1?,IL-6,TNF-?expression decreased;the expression of p16,p21,mTOR,p-mTOR,p70s6 k,p-p70s6 k protein was decreased(P<0.05),and the expression of AMPK,p-AMPK,LKB1,p-LKB1 protein was elevated(P < 0.05).Conclusion: The study showed that high glucose can induce aortic senescence in mice,and the extract of Ginseng,Sanqi and Chuanxiong can improve the aortic senescence induced by high glucose in mice,which may be through the regulation of AMPK/mTOR pathway and its upstream and downstream pathways.
