The dry roots of Disporum cantoniense or Disporum sessile which belonging to the Disporum Salisb and Polygonnateae tribe from Liliaceae family.It is a Commonly used medicinal material in Guangxi,Guizhou and other places.At present,cough syrup and capsules with it as the main drug have been developed and utilized.Its main chemical constituents are polyphenols,flavonoids and their glycosides,fatty acids and nitrogen-containing compounds.These compounds determine that Disporum cantoniense has many effects,including anti-inflammatory.This paper study it the following aspects.First of all,in order to systematically study the chemical constituents of Disporum cantoniense,search for its effective substances,in this experiment,75% ethanol extract was separated by ion exchange chromatography,medium pressure MCI column chromatography,ODS column chromatography,Sephadex LH-20 column chromatography,preparative liquid phase and semi-preparative liquid phase,and a total of 46 compounds were separated.Their structures were identified by ESI-MS,1H-NMR and 13C-NMR techniques.The names of these compounds are as follows: 2-hydroxybenzyl alcohol(A-1),p-hydroxybenzaldehyde(A-2),p-hydroxyacetophenone(A-3),3,5-dimethoxy-4-hydroxy-benzaldehyde(A-4),4-(4-hydroxyphenyl)-2-butanone(A-5),3-hydroxy-1-(4-hydroxy-3,5-dimethoxy-phenyl)-1-propanone(A-6),icariol A2(A-7),neoliquiritin(A-8),2,3,5,4'-tetrahydroxy stilbene-2-?-?-D-glucoside(A-9),(E)-1-(4'-hydroxyphenyl)-but-1-en-3-one(A-10),neoechinulin A(A-11),isoquercitrin(B-1),ecdysterone(B-2),marmesinin(B-3),2?-?-Dglucopyranosyloxybenzyl 6-?-L-rhamnopyranosyloxy-2-hydroxy-3-methoxybenzoate(B-4),4'-O-?-D-glucosyl-5-O-methylvisamminol(B-5),glansreginic acid(B-6),hesperidin(B-7),ononin(B-8),nodakenetin(B-9),6-(2-hydroxybenzylamino)-9-beta-D-ribofuranosylpurine(B-10),isorhamnetin-3-O-?-D-glucoside(B-11),quercetin(B-12),2-aminopyridine(B-13),(E)-4-(4-hydroxy-3-methoxyphenyl)but-3-en-2-one(B-14),tenuifoliside A(B-15),(-)-secoisolariciresinol(B-16),7-O-[2?-O-?-Dgalactopyranosyl]-?-Larabinofuranoside(B-17),neosakuranin(B-18),palmatine(C-1),berberine(C-2),2?-?-D-glucopyranosyloxybenzyl 6-?-L-(4?-O-acetyl)rhamnopyranosyloxy-2-hydroxy-3-methoxybenzoate(C-3),dihydroxyisoechinulin A(C-4),icariin(C-5),torachrysone-8-O-?-D-glucoside(C-6),luteolin(C-7),4',7-Dihydroxyflavone(C-8),epimedin C(D-1),4-hexenoic acid(D-2),emodin-8-Oglucoside(D-3),(2S)-2,3-dihydroxypropyl-1,6,8-tridihydroxy-3-methyl-9,10-dioxoanthracene-2-carboxylate(D-4),nobiletin(E-1),7,8-dehydro-2-hydroxycalamenene-14-ol(E-2),dibutyl phthalate(E-3),2''-O-rhamnosylicariside II(E-4),icariside II(E-5).In the middle of them,compound C-3 is a new compound.Compounds A-1,A-3~A-11,B-2,B-3,B-5~B-11,B-13~B-18,C-1,C-2,C-4~C-6,C-8,D-1~D-4,E-1,E-2,E-4,E-5(a total of 39)is obtain from Disporum Salisb for the first time.Secondly,in order to determine the anti-inflammatory components of Disporum cantoniense,the traditional water extraction and alcohol precipitation method was used to extract the polysaccharide of Disporum cantoniense.Three polysaccharides flow fraction T1~T3 was separated by macroporous resin column and anion exchange column chromatography.The content of polysaccharide and protein in crude polysaccharide was determined by method of phenol-sulfuric acid and Coomassie brilliant blue.Finally,on the basis of chemical research work,combined with the basic activity of Disporum cantoniense and the existing experimental conditions,some of the isolated compounds and polysaccharide flow fraction were screened.The inflammatory model of mouse macrophage RAW264.7 induced by LPS was used,and the content of NO was determined by Griess reagent to judge the anti-inflammatory activity of compounds and polysaccharides.The results showed that: Compounds A-1,A-2,A-4,A-5,A-11,B-4,B-12,B-18,C-7,C-3,D-4,E-1 and polysaccharide flow fraction T1 could inhibit the production of NO without obvious cytotoxicity.
