Compatibility is one of the traditional Chinese medicine attenuating methods.Traditional Chinese medicine compatibility is a common form of traditional Chinese medicine.Its compatibility has always been a hot topic,but because of its complexity,it has not made a big breakthrough,and the drug is used as a traditional Chinese medicine.The core of the compound composition reflects the efficacy of the classic compound,making the series compound have something in common.Therefore,the study of the compatibility of drugs can be used as an important entry point to study the compatibility of the compound,thus revealing its compatibility and scientific connotation,deepening and improving the compatibility theory of traditional Chinese medicine compound,providing clues and basis for the study of the compatibility mechanism of the compound.At the same time,it provides research ideas for the development of new Chinese medicine based on medicine.Therefore,this article will examine the mechanism of compatibility of drugs,and provide reference for the in-depth study of the compatibility of drugs in the future.The use of Astragalus as a model for replenishing qi and invigorating spleen,Monkshood as a toxic tool,and research on Astragalus,Aconiti Lateralis Radix Preparata and their compatibility Absorption characteristics and absorption mechanism of Monkshood alkaloids,and three drug efflux transporters?P-gp,Mrp2,Bcrp?and three tight junction proteins?Claudin-1,Occludin,Zo-1?against Caco-2cells and normal rats,changes in impact.1.MTT assay for the cytotoxicity of Astragalus membranaceus,Monkshood and their compatibility on Caco-2 cellsCaco-2 cell model was used to screen the effective concentration dose by MTT method.Under the condition of complete culture medium,the safe effective concentration of Monkshood was 25.41?g·mL-1 at 24 hours of cell action,and in the condition of HBSS culture,the safe concentration of Monkshood was in the range of19.23-1195.36?g·mL-11 at 2 hours of cell action.The combination of Astragalus membranaceus and Monkshood significantly increased the cell survival rate,and the detoxification effect was the best when the cells were treated for 24 hours.2.The effect of Astragalus membranaceus on the absorption of Monkshood and the absorption mechanism of six alkaloids in Monkshood were studied by Transwell transport model of Caco-2 cells.The effects of compatibility of Astragalus membranaceus and Monkshood on the absorption of six alkaloids in Caco-2 cell model showed that the extracts of Astragalus membranaceus from 299.33,598.66 and 1197.32?g·mL-1 inhibited the Papp of Monkshood alkaloids to varying degrees.The absorption mechanism of six alkaloids in Monkshood showed that there might be an active transport process.Statistical analysis was carried out by using MATLAB fitting software.It was found that the absorption and transport of six alkaloids in Monkshood at the cellular level included simple diffusion,passive transport and carrier-mediated transport,and the three efflux transporters of Mrp2,P-gp and Bcrp all participated in the process.Moving transshipment process.3.Western blot and qRT-PCR study on the effects of Astragalus,Monkshood and their compatibility on three efflux transporters and three tight junction proteins in Caco-2cellsWestern blot analysis showed that the drug transporter?P-gp,Mrp2,Bcrp?and tight junction protein?Claudin-1,Occludin,Zo-1?increased in Caco-2 cells to different degrees,compared with the normal group.P-gp and Mrp2 at low,medium and high doses and Bcrp at high doses were significant?P<0.05?;Monkshood significantly down-regulated the expression of Mrp2,and there was no statistical difference in the expression of P-gp,which was significantly up-regulated the expression of Bcrp;after the combination of Astragalus and Monkshood,the expression of the three proteins was up-regulated,and the three compatibility ratios up-regulated the expression of Mrp2 protein,but there was no statistical difference.The compatibility ratio was 1:1 and 1:2 P-gp and Bcrp protein expression showed significant up-regulation?P<0.05?.The expression of Claudin-1 and Occludin protein was significantly up-regulated with the increase of jaundice concentration?P<0.05?.The expression of Zo-1 protein was significantly up-regulated at low and medium doses,while the expression was significantly down-regulated at high dose?P<0.05?;Zo-1 and Claudin-1 proteins were significantly up-regulated under the action of Monkshood?P<0.05?,while the expression of Occludin protein was not different.The combination of Astragalus and Monkshood promoted the expression of Zo-1 and Claudin-1 proteins.The ratios of 1:1 and 1:2 were significantly up-regulated?P<0.05?;while Occludin protein was significantly down-regulated at these two ratios?P<0.05?,and significantly up-regulated at a ratio of 1:1/2?P<0.05?.<0.05).From this we can infer that the compatibility of Astragalus and Monkshood has the effect of reducing auxin,and three kinds of efflux proteins are involved in the transport of six alkaloids of Monkshood,and that Astragalus can regulate efflux proteins?P-gp,Mrp2,Bcrp?.And the expression of tight junction proteins?Claudin-1,Occludin,ZO-1?,reducing the absorption of Monkshood components,suggesting that Astragalus and Monkshood can affect the transport of the combination by regulating the expression of these transporters and tight junction proteins.Other clinical medications offer new directions and new guidance.4.Western blot and qRT-PCR study of the effects of Astragalus,Monkshood and their compatibility on the expression of three efflux transporters and three tight junction proteins in normal ratsUsing in vitro and in vivo models to prove that Astragalus can inhibit the absorption of Monkshood by regulating the expression of three efflux transporters and three tight junction proteins,so 48 normal rats were intragastrically administered,western blot and qRT-PCR results It is basically consistent with the results of cell experiments.5.Immunofluorescence experiment to study the effects of Astragalus,Monkshood and their compatibility on the expression,location and morphological changes of three tight junction proteins in normal ratsThe results of immunofluorescence experiments confirmed that Astragalus membranaceus can affect the absorption of substances by regulating the expression of three tight junction proteins.It is also suggested that Astragalus may inhibit the absorption of harmful substances by regulating tight junctions and/or promoting the secretion of goblet cells.6.Western blot and qRT-PCR study of the effects of Astragalus,Monkshood and their compatibility on the expression of three efflux transporters and three tight junction proteins in rats with spleen deficiencyTo investigate whether the effects of Astragalus and Monkshood on the expression and transcription of three efflux proteins and three tight junction proteins were different in different functional states.The results showed that the rats had dysfunction of physiological function,jaundice,Monkshood and Its compatibility has altered the expression and transcription of both in vitro and integrin proteins and tight junction proteins,and the translation and transcription levels of these proteins are not consistent.
