Chronic unpredictable mild stress(CUMS)can stimulate the hypothalamic pituitary adrenal axis to over-release glucocorticoids,trigger inflammation,reduce neurogenesis,and induce dysfunction of neurotransmitter systems,which all occur in depression.Seahorses were reported have multiple biological and medical effects,including anti-aging,anti-fatigue,antioxidants,and neuroprotection.However,the mechanism by which seahorses benefit mental health is unclear.The present study systematically evaluate the antidepressant effect of seahorse on mice model induced by CUMS,from neuroendocrine to neuroinflammation,then to neurotrophin and neurotransmitters.Methods:(1)In this study,the depression model is set up by CUMS,which last for 8 week,with the intervention treatment of seahorse dry powder at a concentration of 0.22%?(2)Elevated plus maze,open field test,forced swim test,social interaction test and sucrose preference test are used to evaluate depression-like and anxiety-like behavior.(3)Serum corticosterone,hippocampal IL-1? concentration,and the concentrations of IL-1? and IL-10 as well as activities/content of SOD,GSH-P_x and ROS in the amygdala were measured by ELISA kits.(4)The expression of CD11b,GFAP and BDNF,as well as TrkB expression are evaluated by Western Blot.(5)The content of 5-HT,NA and D,as well as metabolite MHPG,5-HIAA,DOPAC and HVA are detected by HPLC.Results:(1)Compared with the control,the CUMS model mice showed declined in sucrose preference,reduced the exploration ability in the open field,and decreased the exploration and communication ability in the social behavior test,and less number and time of entering in the open arm of the elevated plus maze.While the increased immobility time in forced swim test.After the intervention treatment of seahorse,the sucrose preference has been greatly improved,exploration ability in the open field,the elevated plus maze test and social behavior test are all normalized to standard level,the same as forced swim test.(2)Compared with the control,serum corticosterone,hippocampal IL-1? concentration in the CUMS model mice are dramatically increased,the same as IL-1? concentration in amygdala,while IL-10 level is decreased.Additionally,the activities/content of ROS is significantly elevated in amygdala,while SOD and GSH-P_x are declined.After the intervention treatment of seahorse,serum corticosterone and hippocampal IL-1 concentration are both normalized to standard levels,and IL-1 concentration in amygdala is remarkably decreased,while IL-10 increased.Moreover,activities of SOD,GSH-P_x and ROS in the amygdala are also reversed to standard levels.(3)Compared with the control,the protein expression of CD11b and TrkB is remarkably elevated in hippocampus,while GFAP and BDNF decreased.After the intervention treatment of seahorse,CD11b,TrkB,GFAP and BDNF are all normalized to standard levels.(4)Compared with the control,the concentrations of NA,5-HT and DA are significantly decreased,as well as its metabolite,NA/MHPG,5-HT/5-HIAA,DA/DOPAC and DA/HVA are also decreased.After the treatment of seahorse,all are normalized to standard levels.Conculsion:(1)CUMS can induce depressive and anxiety-like behaviors in mice,which lead to neuroendocrine disorder,neuroinflammation and oxidative stress,and decreased in neurotrophion and neurotransmitters.(2)Seahorse,a natural Marine drug,is likely to improve anxiety and depression-like behavior by reducing oxidative stress and inflammation and regulating the function of neurotransmitters and neurotrophin.
