Objective 1.To investigate the relationship between tumor stem cells and biological behavior of ameloblastoma(AM)by detecting the expression of P75 NTR and SOX2 in ameloblasts;Method 1.Clinical data: The specimens were obtained from 21 patients with ameloblastoma who were admitted to the Department of Stomatology,Affiliated Hospital of Inner Mongolia Medical University from January 2015 to December 2018.All cases had complete clinical and pathological data.There were 9 males and 12 females.Among them,there were 3 cases of maxillary bone and 18 cases of mandible.The 5 cases of dentigerous cysts(DC)as controls.2.Methods: Immunohistochemical method was used and immunohistochemical staining was performed according to SP method.,3.Analysis of immunohistochemistry results: In P75 NTR staining group,the cytoplasm was brown-yellow positive,while in SOX2 staining group,the nucleus was brown-yellow positive.4.Statistical analysis: The results were analyzed by SPSS 22.0 statistical software.The differences of P75 NTR and SOX2 expression in solid ameloblastoma and single cystic ameloblastoma were compared with those in central and peripheral positive cells in solid ameloblastoma.Kolmogorov Smirnov test was used to detect the difference of Lia=0.05.When the test standard was P < 0.05,the difference was statistically significant.Result:The expression of P75NTR:(1)In the solid AM,the expression of P75 NTR was observed in each case,and the expression of each subtype was different.In follicular,plexiform,and acanthoma AM,The epithelial cell membrane and cytoplasm are strongly expressed in peripheral columnar cells,while the central star reticular epithelial cells are weakly expressed or no expressed.Almost all cell membranes and cytoplasm in epithelial islands of basal cell type AM were strongly expressed.(2)In 5 cases of unicystic AM,there is no type II AM,wall type meloblastoma,which are mixed type I and type III,and the expression pattern of type I unicystic AM is similar with the dentiguious cyst,and the type III unicystic AM is simler with the multicystic AM.(3)In 6 cases of dentigerous cysts,P75 NTR expression is observed in all cases,which are strong expressed in the cytoplasm of epithelial cells close to the basement membrane,while epithelial cells away from the basement membrane are no expressed.The expression intensity and expression rate of P75 NTR in the surrounding columnar cells were significantly higher than those in the central reticular cells,P<0.05,and the difference is statistically significant.Compared with dentigerous cysts,the expression rate and expression intensity of cells in ameloblastoma the difference is not statistically significant.Expression of SOX2: SOX2 was not expressed in follicular,acanthoma,and basal cell,unicystic ameloblastoma.In 5 cases of plexiform ameloblastoma,1 case of epithelial cells showed moderate to strong expression in the nucleus;in 6 cases of dentigerous cysts,1 case of epithelial cells adjacent to the basement membrane is strongly expressed,while the cells away from the basement membrane is moderate expressed.Compared with dentigerous cysts,the cell expression rate and expression intensity of Sox2 in ameloblastoma has no different,P>0.05,and the difference was not statistically significant.Conclusion: In ameloblastoma,the expression of the stem cell marker P75 NTR is expressed in the columnar epithelial cells adjacent to the basement membrane.Considering its correlation with the proliferation of tumor cells,SOX2 is expressed as a stem cell marker in various malignant tumors,but the expression in ameloblastoma is rear,and its role in ameloblastoma needs further investigation.
