Objective To study the combination of the single component extracts of Sen Deng-4 and its disassembled group on vascular endothelial growth factor(VEGF)in serum and synovial membrane of collagen-induced arthritis rats(CIA).The effects of angiosin(Ang)content and expression on the neovascularization of collagen-induced arthritis rats were systematically analyzed.Methods Rat arthritis model was established by bovine type II collagen induction method,and the rats were divided into normal group(distilled water),model group(distilled water),tripterygium glycosides tablets(9.45 mg·kg-1 dose),Mori Deng-4 soup low dose group(1.35g·kg-1·d-1 dose),Sendeng-4 soup medium dose group(2.7g·kg-1·d-1 dose),Sendeng-4 high dose Group(5.4g·kg-1·d-1 dose),Wenguanmu extract group(19.14mg·kg-1·d-1 dose),and medlar extract group(8.86mg·kg-1·d-1 dose),hazelnut extract group(15.97mg·kg-1·d-1 dose),Chuanxiongzi extract group(56.9mg·kg-1·d-1 dose),continuous administration for 4 weeks.The pathological morphology and neovascular density of synovial tissue were observed after HE staining in rat ankle joint and knee joint.The contents of VEGF/VEGFR,Ang1,Ang2 and Tie2 in rat serum were determined by enzyme-linked immunosorbent assay(ELISA).Western blotting(Western Blot)was used to detect the expression of VEGF/VEGFR,Ang1,Ang2 and Tie2 in rat synovial tissue.Results The results of HE staining showed that a large number of exuded inflammatory cells were observed in the ankle joint of the model control group,synovial cell hyperplasia,neovascularization increased significantly and vasospasm was formed.The neovascular density of each treatment group was different from that of the model group.The reduction was significantly improved in the Tripterygium glycosides group and the Senden-4 high-dose group,and the decomposed experiment was more effective in the Wenguanmu extract group.The results of Elisa showed that the levels ofserum VEGF/VEGFR,Ang1,Ang2 and Tie2 in the model group were significantly higher than those in the normal group(P<0.05).The positive expression of each group was lower than that in the model group(P<0.05).Among them,the decline of the high dose group of Tripterygium wilfordii and Senden-4 was the most significant,and the decline of the Chinese Guanmu group in the decomposed experiment was more obvious than the other groups.The results of Western Blot showed that VEGF/VEGFR,Ang1,Ang2 and Tie2 protein were expressed in all groups.Compared with the normal group,VEGF,Ang1 and Ang2 proteins were highly expressed in the model group and each drug group(P<0.05).Compared with the model control group,the expressions of VEGF,Ang1,and Ang2 protein in the tripterygium glycosides group,the Sendeng-4 dose group and the Wenguanmu group were decreased(P<0.05),while the Sendeng-4 high dose group was compared.There was no significant difference in histone expression between Tripterygium wilfordii polysaccharides(P>0.05).There was no significant difference between VEGFR and Tie2 protein in normal group and each model group(P>0.05).Conclusion Mongolian Medicine Senden-4 can effectively reduce the expression of vascular endothelial growth factor and angiogenin in serum and protein of collagen-induced arthritis rats,and then reduce the degree of bone destruction by reducing the formation of vasospasm.-4 provides a theoretical basis for the treatment of rheumatoid arthritis.The Chinese crown wood of each split group is more effective than the other three single-flavor drugs,indicating that Wenguanmu may be the main drug for the action of Senden-4.Therefore,it is worthwhile to further study the Wenguanmu in the future,in order to clarify the treatment of Mongolian medicine.New targets and extensions for rheumatoid arthritis have found clinically effective new botanicals.
