| Objective: 1.By observing the effect of different acupuncture methods of Mongolian medicine on the inquiry behavior and the level of lack of pleasure in depression rats,discussing the mechanism of the treatment of depression with different acupuncture methods of Mongolian medicine.2.By observing monoamine neurotransmitters and oxidative stress in hippocampus and frontal lobe of noradrenaline(NE),dopamine(DA),5-hydroxytryptamine(5-HT),and oxidative stress Correlation between the changes of superoxide dismutase(SOD)and ketone microalbuminaldehyde(MDA),and the effects of different acupuncture methods on neurotransmitters in hippocampus and frontal lobe of depressed rats and antioxidant mechanism.METHODS: Male Sprague-Dawley rats were randomly divided into blank group,model group,Mongolian medicine silver needle group,Mongolian medicine warm needle group,drug group and acupuncture combined group,with 8 rats in each group.All rats in the other groups in the blank group were orphaned,and 7 different stress stimuli were randomly received for 28 days.The rats were stimulated daily,and the same kind of stimulation could not occur continuously,and each stimulus should appear at least 3 times.Blank group: Let the rats drink water normally,ingest food,and do not give any stimulation.Model group: isolated,and received a variety of unpredictable stress stimuli for 28 d.Drug group: 1 hour before the daily stress stimulation,Prozac was formulated into 1 mg/1 ml with physiological saline,and administered orally according to 2 mg/kg body weight.Mongolian medical silver needle group: 1 hour before receiving various kinds of stress stimulation,the Mongolian medical silver needle stimulates the Badakan acupoint,heart point and black and white points.Each time you need to leave the needle for 20 min.Mongolian medicine warm needle method: acupuncture and acupoints are the same as the Mongolian medical silver needle group.Take one acupuncture point every day.Needle bing should be connected with Mongolian medical YY-I type electric hot silver needle therapeutic instrument for heating stimulation,each heating for 20 min.To prevent burns on the acupuncture site.The heating step was the first day of the Bada dry hole,the second day was the heart point,and the third day was the black and white hole,and the rats were sequentially stimulated in this order.Acupuncture and medicine combination group: isolated,1 hour before daily stress stimulation,the same method of Mongolian medicine warm needle group stimulation,after the stimulation,Prozac was formulated into 1mg/1ml with normal saline,according to 2mg/kg body weight,administered by intragastric administration.Each group of treatment methods was 28 days.Rats' behavior was measured within 24 hours after 28 days using open-field open-end experiments and sucrose consumption,followed by decapitation,hippocampus and prefrontal cortex,and LC,MS,and MS were used to detect NE,DA,and 5-HT.Expression,SOD activity and MDA content were determined by xanthine oxidase method and thiobarbituric acid method.Results: 1.Observing the influence of different acupuncture methods on the behavior of depressed rats,the consumption of sugar water in Mongolian medicine Yinzhen group,Mongolian medicine warm needle group and acupuncture combined group was less than that of blank group,but the model group Significantly elevated(P <0.05).The consumption of sugar water indicates that different acupuncture methods of Mongolian medicine have a significant effect on the pleasure of depression model.In the open-field open-end experiment,the horizontal and vertical movements of the Mongolian medical silver needle group,the Mongolian medicine warm needle group,and the acupuncture-medicine combination group were reduced compared with the blank group,but significantly higher than the model group(P< 0.05).It shows the influence of different acupuncture methods of Mongolian medicine on the exploration ability of depressed rats.2.The expression and SOD activity of NE,DA and 5-HT in the hippocampus and frontal lobe of the model group were significantly lower than those in the blank group(P<0.01).Compared with the model group,the silver needle group of Mongolian medicine,the warm needle group of Mongolian medicine,the drug group and The expressions of NE,DA,5-HT and SOD in the hippocampus and frontal lobe of the acupuncture-medicine combination group were increased(P<0.05).Compared with the blank group,the MDA of hippocampus and frontal lobe in the model group was significantly increased(P<0.01).Compared with the model group,the MDA of the frontal lobe of the Mongolian medicine warm needle group,the drug group and the acupuncture combined group was significantly lower(P<0.05).).Conclusions: 1.Through behavioral testing,different acupuncture methods of Mongolian medicine can improve the core symptoms such as exploration ability and loss of pleasure in depressed rats.2.Different acupuncture methods of Mongolian medicine can improve the core symptoms of depression,and its mechanism may be related to the improvement of NE,DA,5-HT expression and SOD activity,and the reduction of MDA. |
