Objective To investigate the effect of Mongolian medicine Danggong-3 on atherosclerosis(AS)rats and its possible mechanism.Methods The AS rat model was established by high-fat diet combined with vitamin D3 injection.64 male Wistar rats were divided into normal group and model group.The successful atherosclerotic rats were randomly divided into Danggong-3 high,medium and low dose groups,Gemfibrozil group and model control,10 for each group.After successful modeling,continuous administration for 4 weeks,the blank control group and the model control group were given the same volume of distilled water.After 4 weeks of administration,the morphological changes of blood lipids and aorta in each group were observed,and the levels of matrix metalloproteinase(MMP-2),matrix metalloproteinase-9(MMP-9)and nuclear factor(NF-κB)were detected.Results 1.In terms of blood lipid level,Danggong-3 low,medium,high dose group and Gemfibrozil group could reduce TC,TG,LDL-C levels and increase HDL-C levels in AS rats(P< 0.05).2.Under light microscope,the pathological damage of aorta in the low,middle and high doses of Danggong-3 and the Gemfibrozil group was significantly lighter than the model control group;3.Compared with the blank control group,the levels of MMP-2 and MMP-9 in the model group,Danggong-3 low,medium and high dose group and Gemfibrozil group increased significantly(P< 0.05),while the levels of NF-κB did not change significantly(P<0.05).Compared with the model control group,the levels of MMP-2 and MMP-9 in Danggong-3 low,medium and high dose group and Gemfibrozil group decreased significantly(P<0.05),while the levels of NF-κB did not change significantly(P< 0.05).Conclusion 1.Danggong-3 significantly down-regulated the levels of TC,TG and LDL-C,and up-regulated the levels of HDL-C in AS rats,It indicated that Danggong-3 could intervene in the development of AS by regulating blood lipid and stabilizing plaque.2.Danggong-3 may improve atherosclerotic plaque by lowering the levels of MMP-2 and MMP-9.3.The effect of Danggong-3 on improving atherosclerotic plaque may not be related to regulating the level of NF-kB.
