Objective: To investigate the role of RNA interference in the knockdown of vimentin,expression in human glioma cells and its biological effects on cell proliferation,invasion and metastasis,as well as targeted gene therapy for glioma diseases in the future.The aspect provides the corresponding theoretical basis.Methods:First,the eukaryotic expression vector of VIM interfering RNA was synthesized by siRNA design principle and confirmed by sequencing.The successfully synthesized eukaryotic expression vector was transfected into the glioma U251 cell line by liposome method in cell experiments.RT-qPCR was used to detect the mRNA expression of VIM in U251 cells after VIM RNAi interference.The vimentin interference group was set as the experimental group,and the control group was the transfected siRNA negative control sequence group.Finally,MTT method,cell scratch test and Transwell chamber invasion test were used to observe the changes of tumor cell growth,invasion and metastasis after interference.The experimental results were compared and some conclusions were drawn.Resalts:1.The VIM interfering RNA eukaryotic expression vector was successfully synthesized and tested into the human glioma cells cultured by the liposome method.2.One group transfected with VIM RNAi was established as an experimental group,and the control group was a group transfected with a negative control sequence.3.MTT results showed that compared with the control group,the cell proliferation ability of the experimental group decreased,which was statistically significant(P<0.05).The transwell chamber invasion test showed that the experimental group had lower invasive ability and statistical significance(P<0.05);and using the cell scratch test,the cell transfer ability of the experimental group was relatively weakened,which was statistically significant(P<0.05).Conclusion: The VIM interfering RNA eukaryotic expression vector was successfully synthesized by RNA interference technology,which inhibited the expression of vimentin.The biological capacity of human glioma cells after growth inhibition,invasion and metastasis was reduced compared with the control group.It can be seen that the deeper exploration of the application of RNAi technology to knock out vimentin provides new ideas for the treatment of human glioma in the future.
