Objective Invasion and migration are the main causes of death in colorectal cancer,and proliferation also affects the degree of malignancy,but the mechanism remains unclear.Micro RNA(mi RNA)is a small non-coding RNA molecule that is involved in the regulation of gene silencing and cancer progression,and also affects cellular functions including malignant transformation,angiogenesis,cell growth and inflammatory responses.Mir-375 is a multifunctional mi RNA involved in islet development,glucose homeostasis,mucosal immunity,lung surfactant secretion,and tumorigenesis.Notch signaling pathway is an evolutionarily conserved cellular signaling pathway,which has an impact on cancer occurrence,cancer stem cell biology and tumor angiogenesis,and these effects determine its important status as a potential therapeutic target in the field of oncology.Notch signaling pathway is considered to play a role in a variety of cancers and is a necessary condition for regulating cell lineage differentiation of normal intestinal mucosa.Notch2 expression is closely related to CRC and may play a role in tumor inhibition.In this study,the factors influencing the invasion,migration and proliferation of colorectal cancer were studied from the molecular level,and the effect of microrna-375(mir-375)on the invasion,migration and proliferation of colorectal cancer(CRC)and its relationship with the Notch2 pathway protein were discussed.Methods The lentivirus carrying mir-375 was transfected into human colorectal cancer cells HCT-116 as the experimental group,the null lentivirus was transfected into HCT-116 cells as the negative control group(NC group),and the untransfected HCT-116 cells as the blank group.Quantitative Real-time PCR(qrt-pcr)was used to test the transfection efficiency,Transwell test were used to evaluate cell invasion and migration abilities,CCK-8 assay was used to detect cell proliferation and Western Blot was used to detect the change of Notch2 pathway protein expression.The Notch2 small interfering RNA(si RNA)was transfected into HCT-116 cells as the notch2-sirna group,the negative control was transfected into HCT-116 cells as the Notch2 negative control group,and the untransfected HCT-116 cells as the blank group.Western Blot was used to detect the expression of Notch2 protein in the cells of the three groups,the effect of Notch2 changes on invasion and migration of colorectal cancer cell was further verified by Transwell test,and Cck-8 assay was used to detect cell proliferation.Results Real-time fluorescence quantitative PCR showed that the expression of mir-375 in colorectal cancer tissues and cells were significantly lower than that in human normal colorectal mucosa cells FHC(P<0.01,P<0.001).Compared with the NC group and the blank group,in the Transwell chamber experiment,the invasion and migration abilities of cells in the experimental group were decreased(P<0.01),in the scratch test,the migration ability of cells in the experimental group were decreased(P<0.001),in the cck-8 experiment,the proliferation ability of cells in the experimental group was decreased(P< 0.05)and the relative expression level of Notch2 pathway protein in cells in the experimental group was decreased showed by Western Blot(P<0.01).Notch2 was down-regulated in HCT-116 cells.Compared with the Notch2 negative control group and the blank group,the invasion and migration abilities of cells in the notch2-sirna group were decreased in the Transwell chamber experiment(P<0.01),and the proliferation ability of cells in the notch2-sirna group was decreased in the cck-8 experiment(P<0.05).The differences were statistically significant.Conclusion Mir-375 can inhibit the expression of the Notch2 pathway protein,thereby inhibiting the invasion,migration and proliferation of colorectal cancer cells.When Notch2 protein is down-regulated with small interfering RNA,the invasion,migration and proliferation ability of colorectal cancer cells is decreased.
