Objective: To observe the effect of anshen buxin pill on the learning ability of normal mice with memory acquisition,consolidation and reproduction disorder and the transgenic model of APP/PS1,in order to prove that the drug can improve memory.Methods: 1.Pathological model of memory acquisition,consolidation and reproduction disorders in normal mice was designed.Randomly divided into normal control group,model group,anshen buxin pill high(1.56 g/kg),medium(0.78 g/kg),low(0.39 g/kg)dose group and positive control drug haberin group(huperzine a,0.05 mg/kg).In addition to the normal control group and the model group,the other groups were given corresponding drugs to gavage,once a day for 12 days.On day 13,normal control group was intraperitoneally injected with normal saline,and the rest groups were injected with modeling drugs respectively.After 24 hours,the jumping platform method and Morris water maze method(only used for scopolamine disorder model)were used to test the learning performance of the mice.2.Experimental design of APP/PS1 pathological model: 64 mice were randomly divided into6 groups,namely,negative control group(C57,11 mice),model group(APP/PS1,11 mice),anshen buxin pill high(APP/PS1,10 mice),medium(APP/PS1,11 mice),low(APP/PS1,10mice)dose group and haberin group(huperzine a,11 mice),dose was the same as above.In addition to the negative control group and model group of distilled water to fill the stomach once A day,other groups are all made of the corresponding drug lavage,11 weeks continuously,with MT-200 type Morris water maze test grades groups of mice,and then enzymatic SOD and MDA in the serum and brain tissue of mice was detected in acetyl cholinesterase activity,HE dyeing hippocampus histopathologic,observing A beta immunohistochemical CA1 area brain plaque deposits.All the experimental results were analyzed by SPSS 17.0 statistical software.Results: 1.Compared with the normal group,the diving incubation period of mice treated with scopolamine hydrobromide,sodium nitrite and ethanol was significantly shortened,and the number of errors was significantly increased(p<0.01).Compared with the model groups, the diving incubation period of anshen buxin pill in medium and high dose groups was significantly prolonged,and the number of errors was significantly reduced(p<0.05 or p<0.01).In Morris water maze navigation test,compared with the control group,the scopolamine model group showed significantly longer swimming incubation period and swimming distance(p<0.05 or p<0.01),indicating the successful replication of the model.Compared with the model group,the incubation period and swimming distance of anshen buxin pill were significantly shortened in the medium and high dose groups(p<0.05).In the space exploration experiment,compared with the control group,the scopolamine model group had shorter swimming time in the target quadrant(p<0.05),longer time to reach the platform at the first time(p<0.01),and significantly fewer shuttle times(p<0.01).Compared with the model group,the swimming time of mice in the target quadrant of anshen buxin pill in medium and high dose groups was significantly prolonged(p<0.05),the time to reach the platform for the first time was shortened(p<0.05),and the number of shuttle was increased(p<0.05).2.There was no statistically significant difference in weight gain in each group(p>0.05).In the directional navigation experiment of water maze,compared with the negative control group,the escape incubation period and swimming distance of the model group mice were significantly prolonged(p<0.05),indicating that the model group mice had learning and memory impairment.Compared with the model group,the escape latency and swimming distance of anshen buxin pill in low,medium and high dose groups and haberin groups were significantly shortened,and the difference between the medium and high dose groups was statistically significant(p<0.05).In the space exploration experiment,compared with the negative control group,the swimming time in the target quadrant of the model group was significantly shortened(p<0.05),and the shuttling times were significantly reduced(p<0.01).Compared with the model group,the swimming time in the target quadrant of anshen buxin pill group,high dose group and haberin group was significantly prolonged(p<0.05 or p<0.01),and the shuttling times were increased(p<0.01).Compared with control group,model group mice serum MDA level significantly increased(p<0.01),SOD activity decreased obviously(p<0.01),acetylcholinesterase(ACh E)activity increased significantly(p<0.01),disordered arrangement of hippocampal CA1 area cells,cells with narrow,interstitial edema of osteoporosis,nissl body to reduce or disappear,A beta 40 positive cells increased significantly(p<0.05),the average grey value dyeing deepen(p<0.05).Compared with the model group,the MDA level in anshen buxin pill group,high dose group and haberin group was significantly reduced(p<0.05),and SOD activity was significantly increased(p<0.05).ACh E activity was significantly reduced(p<0.05).Under the microscope of HE staining,we could see that the cells in hippocampal CA1 area were well arranged,most of them had obvious nucleoli,clear cell membrane and abundant chromatin,some cells were pyretic,the staining was deepened,the volume was reduced,and some interstitium was loose and edema.A beta 40 positive cell count,anshen buxin pill low and medium dose groups showed A trend of reduction,but the difference was not statistically significant(p>0.05).Mice in the high-dose group and haberin group showed the most significant decrease(p<0.05),and the mean gray value of positive cells in the low-dose group and the medium-dose group showed no difference compared with the model group(p>0.05),while the mean gray value and staining became lighter in the high-dose group and haberin group significantly increased(p<0.05).Conclusions: Anshen buxin pill is effective in the memory impairment model of normal mice induced by scopolamine hydrobromide,sodium nitrite and ethanol in clinical dose and high dose.At the same time,the drug can also improve the learning and memory ability of APP/PS1 transgenic senile dementia model mice in medium and high dose,indicating that anshen buxin pill has beneficial effects on intelligence.
