Objectives To investigate the pretreatment of escitalopram(ESC)for acute Parkinson's disease(PD)caused by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP).The effects of motor function in model mice.Methods Thirty-seven clean healthy male C57BL/6 mice at 23 weeks of age and 23 to30 g were randomly divided into 3 groups: normal control group(CON group),acute PD model group(PD group),and ESC pretreatment group(ESC group).),the latter two groups were made into an acute PD model.2 The model was successfully produced after the elevated cross maze experimental observation inde(?)was the percentage of the number of times the mouse entered the open arm to the total number of arms(OE%)and the open arm stay time as a percentage of the total arm time(OT%)to detect the mouse Anxiety changes;the total distance of free movement of the mice,the retention time of the central area and the number of feces were used to test the presence or absence of depression in the mine.The number of physical crossings,the number of vertical uprights and the number of grooming were observed.Check the motor function of the mouse.3 Real-time fluorescence quantitative(RT-PCR)assay was used to detect the changes of brain-derived neurotrophic factor(BDNF)mRNA in hippocampus and striatum of mice.4 Using SPSS22.0 software system for statistical analysis of data,measurement data for homogeneity analysis of variance,using mean ± standard deviation((?)± s),LSD test for variance homogeneity data,Dunnett test for data with irregular variance The significant standard was bilaterally?=0.05,and the difference was statistically significant at P<0.05.Results 1 In the early stage of acute MPTP injection,the mice in PD model showed lack of flexibility,less movement,tremors,stiff symptoms such as difficulty in bending of the spine and limbs,but tremors were not obvious during active exercise,and the movement was slow.The above symptoms were in 24 hours.After gradually weakening or even disappearing.2 The number of open arms in the elevated plus maze test mice accounted for the percentage of total number of arms(OE%): ESC group(46.56 ± 16.96),PD group(47.09±17.73),CON group(38.90±13.02);open arm residence time Percentage of total arm length(OT%): ESC group(30.69 ±21.14),PD group(32.00±21.81),CON group(29.80±20.73),there was no significant difference between the groups(P>0.05).The total free movement distance of the experimental mice in the mine: CON group(263.39 ±60.79),PD group(276.89 ±51.62),ESC group(278.02 ± 84.94);central area retention time: CON group(63.97±30.50),PD group(81.20±42.23),ESC group(60.67±31.11);feces count: CON group(1.50±1.08),PD group(2.00±1.41),ESC group(1.38±1.47),the differences of the above groups were not statistically significant(P>0.05);the number of horizontal crossings: compared with the CON group(288.90±3.03)and the ESC group(291.19±3.90),the PD group(262.36±2.69)mice had significantly reduced the number of crossings,and the differences in each group were different.Statistical significance(P<0.05),but there was no significant difference in the number of crossings between the CON group and the ESC group(P>0.05).The number of body uprights: PD group(57.55±3.72)compared with CON group(89.90±4.58).In the ESC group(91.56±4.45),the number of body erectiles decreased,and the differences were statistically significant(P<0.05).However,there was no significant difference in the number of erectiles between the CON group and the ESC group(P>0.05).Number of times: compared with the CON group(11.60 ± 1.64)and the ESC group(11.56 ± 1.31),the PD group(10.00 ± 1.26)combed the number of hairs Reducing each group differences were statistically significant(P <0.05),but the CON group,ESC treatment group of mice there was no significant hair modified difference number(P>0.05).3 RT-PCR results showed that compared with CON group,the expression of BDNF mRNA in hippocampus and striatum of PD group was significantly decreased.The BDNF mRNA in hippocampus and striatum of ES group was higher than that in PD group.The expression level of the group was significantly increased(P<0.05),but there was no significant difference in the expression of BDNF mRNA between the hippocampus and striatum in the CON group and the ESC group(P>0.05).Conclusions 1 Acute MPTP injection has certain effects on the motor function of normal mice,which can cause the performance of Parkinson's disease such as static tremor,muscle rigidity and decreased exercise.2 ESC pretreatment can improve motor function defects in acute PD model mice.Figure4;Table4;Reference 149.
