Background: Myocardial ischemia-reperfusion injury is a significant problem in clinical cardiology.It refers to a more serious myocardial injury caused by blood recanalization after a period of myocardial ischemia than that caused by vascular occlusion.Spinal cord as the primary afferent and efferent center of cardiac sensory and sympathetic nerve fibers has received increasing attention in recent years in the regulation of myocardial ischemia-reperfusion injury.Previous Studies showed that myocardial ischemia-reperfusion injury was closely related to abnormal expression of lncRNAs in myocardium,but there were few reports on the effect of altered expression of lncRNAs in spinal cord following myocardial ischemia-reperfusion injury.Objective: To explore the expression patterns of lncRNAs in spinal cord after myocardial ischemia-reperfusion injury and its potential role in the early stage of cardiac reperfusion.Methods: Adult male Sprague Dawley rats(weighing 250-300 g)were randomly assigned to two groups: control group and model group.Then myocardial ischemia-reperfusion injury model was performed.After 30 minutes of myocardial ischemia and 2 hours of reperfusion,the upper thoracic spinal cord tissues were immediately dissected and isolated.LncRNAs and mRNAs in spinal cord tissues were screened by transcriptome sequencing technology and the expression of some highly-deregulated mRNAs and lncRNAs was further validated by RT-qPCR.For investigating the molecular functions in the upper thoracic spinal cord tissues during MIRI,GO and KEGG pathway analyses were used to analyze the dysregulated genes.Results: A total of 126 lncRNAs were differentially expressed,in which 41 probe sets were up-regulated and 85 down-regulated;a total of 470 mRNAs were differentially expressed,in which 231 probe sets were up-regulated and 239 down-regulated.The results of RT-qPCR validation studies in spinal cord revealed that 8 lncRNAs(ENSRNOT00000080713,ENSRNOT00000090564,ENSRNOT00000082588,ENSRNO T00000091570,ENSRNOT00000087777,ENSRNOT00000082061,ENSRNOT00000087 028,ENSRNOT00000086475)and 4 mRNAs(Frs3,Nedd4 l,Tep1,Myef2)showed very different patterns.Conclusion: The altered expression of mRNAs and lncRNAs in upper thoracic spinal cord may play a pivotal role in the process of myocardial ischemia-reperfusion injury,which provides a foundation for understanding the roles and mechanism of lncRNAs in the early stage of reperfusion.
