Objective: To investigate the molecular mechanism of NUAK2 inhibition of colorectal cancer development and metastasis.Methods: Utilizing the knockdown and overexpressed stable vectors,we implemented some experiments in colorectal cancer cell lines such as proliferation,migration,invasion,stemness.Then we established the nude mice tumor model to validate the function of NUAK2 alteration in vivo.In addition,we also executed real-time quantitative PCR,Western Blot,co-immunoprecipitation,immunofluorescence staining and other methods to further explore the mechanism of NUAK2 on regulating the migration and invasion in colorectal cancer cells.Results: The ability of migration and invasion in DLD-1 cell line knocked down with NUAK2 was significantly increased compared with the control.The DLD-1 cell line overexpressing NUAK2 was less able to migrate and invade compared with the control;which was verified in LOVO,Caco-2,and HCT116 cell lines.The results were consistent with the DLD-1 cell line.The nude mice metastasis model also demonstrated that reduced expression of NUAK2 can promote the development of metastases.The interaction between PKM2 and NUAK2 was found by IP mass spectrometry analysis and CO-IP assay.The results of qPCR,Western Blot,immunofluorescence staining and cell migration assay showed that NUAK2 interacts with PKM2 to regulate the expression and influence the migration and invasion in colorectal cancer cells.Conclusion: In colorectal cancer,NUAK2 interacts with PKM2 to regulate the migration and invasion of colorectal cancer cells.The down-regulation of NUAK2 and PKM2 can inhibit the progression and metastasis of colorectal tumors.
