Juglans hopeiensis Hu,which is also called Hebei walnut,pecan,wenwan walnut,etc.,belongs to Juglandaceae.J.hopeiensis is mainly distributed in Hebei,Tianjin and Shanxi provinces.So far,studies on chemical constituents and biological activities of Juglans,such as Juglans mandshurica Maxim.and Juglans regia L.have been reported,but none of the phytochemical studies has been carried out on J.hopeiensis.In order to develop and utilize the resources of J.hopeiensis,our group put the study on chemical constituents and biological activities of pericarp of J.hopeiensis systematically.As a result,63 compounds were isolated from the 95%ethanol extract.The compound types are as follows:diarylheptanoid,triterpenoid,naphthone and other types.The chemical structures of the above 63 compounds were identified by a variety of spectral techniques and physicochemical constant determination.Among them,1-10 are novel diarylheptanoids,and 30-31 are novel triterpenoids.In addition,the cytotoxic,neuroprotective and antibacterial activities of some of the isolated monomeric compounds were tested.In the cytotoxic test,we tested the antitumor activity of diarylheptanoids?1-28?and triterpenoids?29-49?.The experimental results showed that triterpenoid 36exhibited moderate cytotoxicity against MCF7 and HT29 with IC500 of 5.25 and 6.01?M,respectively.The tested diarylheptanoids didn't show cytotoxic activity on either MCF7 or HT29 cancer cell lines at 50?M.The hypoxia injury and oxidative damage model of SH-SY5Y?human neuroblastoma cells?were established by using CoCl2 and H2O2,respectively,to investigate the neuroprotective effects of diarylheptanoids?1-28?.The results showed compound 7 exhibited significant neuroprotective effects,and 3-12,15-17,19-21 and23-28 exerted moderate neuroprotective effects against H2O2-induced cell death of SH-SY5Y.Besides,compounds 1,4,5,7,11,12,14,17,18 and 24 showed moderate neuroprotective effects on CoCl2 treated group.Antibacterial activities of diarylheptanoids?1-28?and triterpenoids?29-49?against E.coli,S.aureus and C.albicans were tested.The results showed that the tested compounds had no significant activities.
