Objective:o detect the expression of ?-catenin and matrix metalloproteinase-3(MMP-3)in cartilage of knee osteoarthritis at different stages.To explore their role in the progression of disease in patients with osteoarthritis and its relationship with pathogenesis.Methods:OA group: patients who underwent artificial knee arthroplasty(TKA)due to osteoarthritis,or those who underwent arthroscopic diagnosis and treatment of osteoarthritis,retained 70 cases of discarded articular cartilage during surgery;NOA group: A total of 13 cases of fresh cartilage donated by patients with traumatic cartilage in the knee joint that could not be repositioned in the knee joint,or those who had undergone amputation due to severe limb injuries and complete joints.According to the WOMAC Osteoarthritis Index,the OA group was 18 in the mild OA group,22 in the moderate OA group,and 30 in the severe OA group.The positive cells of ?-catenin and MMP-3 were stained by immunohistochemistry.According to the expression of the two proteins,the relationship between the two proteins and OA was analyzed by software SPSS 22.0.Results: Immunohistochemical staining of ?-catenin: 2 cases of ?-catenin positive expression in the NOA group,the positive rate was 15.38%;in the articular cartilage of the OA group,10 cases were positive in the mild OA group,the positive rate was 55.56%;In the moderate OA group,17 cases,the positive rate was 77.27%,and the severe OA group was 27 cases,the positive rate was 90.00%.MMP-3 immunohistochemical staining results: 1 case of MMP-3 positive expression in NOA group,the positive rate was 7.69%;in group OA group,9 cases of mild OA group were positive,the positive rate was 50.00%;moderate OA group 14 cases were positive,the positive rate was 63.64%,and 25 cases were severe in the severe OA group,the positive rate was 83.33%.Chi-square test showed that the positive expression rate of ?-catenin and MMP-3 in the osteoarthritis group was significantly higher than that in the non-osteoarthritis group(X2=19.053/16.803,P < 0.001).The results of Spearman correlation analysis indicated that the expression levels of ?-catenin and MMP-3 in cartilage tissue increased with the increase of WOMAC osteoarthritis index,showing a significant positive correlation(r=0.753/0.787,P<0.001).The expression levels of ?-catenin and MMP-3 increased with the increase of Mankin score,showing a significant positive correlation(r=0.840/0.774,P<0.001),and ?-catenin and MMP-3 in OA articular cartilage.There was a significant positive correlation between expression intensity(r = 0.891,P < 0.001).Conclusion:(1)The joints ?-catenin and MMP-3 were involved in the destruction of cartilage during the development of OA,and there was a significant positive correlation between the expression of these two proteins in OA cartilage.(2)The expression of ?-catenin and MMP-3 in OA articular cartilage was significantly enhanced compared with NOA group,and the expression intensity of these two proteins increased with the increasing OA.These two proteins can be used as the basis for early diagnosis of OA.
