Objective To study the expression of ubiquitin E3 ligase(MARCH2)in epithelial ovarian cancer and its influence on the biological behavior of ovarian cancer cell lines and its underlying mechanisms.Methods(1)Immunohistochemistry was used to detect the expression of ubiquitin E3 ligase MARCH2 expression in epithelial ovarian cancer tissues,(2)qPCR was used to detect the expression of MARCH2 in ovarian cancer cell lines SKOV3 and A2780 after siRNA knockdown of MARCH2,(3)After the silencing of MARCH2,EdU assay were used to confirm the proliferation,ability of ovarian cancer cells.(4)After the silencing of MARCH2,Transwell assay were used to confirm the invasion ability of ovarian cancer cells.(5)Western blot was used to detect the expression of MARCH2?P62?LC3?PI3K?AKT?mTOR in SKOV3 and A2780 cell lines after silencing MARCH2 gene.Results(1)Epithelial ovarian cancer tissues showed high expression of MARCH2 compared with normal ovarian tissues,and the expression was related with FIGO stage(P<0.01)and was not related to age.(P>0.05).(2)After silencing MARCH2,the expression of MARCH2 in SKOV3 and A2780 cell lines was lower than that in NC group.(3)After silence of MARCH2,the ability of cellular proliferation of SKOV3 and A2780 cell lines were reduced(P<0.05).(4)After silence of MARCH2,the ability of cellular invasion of SKOV3 and A2780 cell lines were reduced(P<0.05).(5)After silencing MARCH2,Western blot analysis showed that P62 expression was up-regulated,and the expression of MARCH2,LC3,PI3 K,P-AKT and P-mTOR was down-regulated(P<0.05).Conclusion Compared with normal ovarian tissue,MARCH2 is up-regulated in epithelial ovarian cancer tissues.Silencing MARCH2 can affect the biological behavior of ovarian cancer cell lines.The mechanism may be related to the inhibition of autophagy by inactivating PI3K-AKT-mTOR pathway after silencing MARCH2,suggesting that MARCH2 may be a candidate oncogene for ovarian cancer,It might also be a potential target for ovarian cancer treatment.
