Methylation Of PAX-1 Using To HPV Positive Patients For Screening Different Lesion

Among worldwide woman,cervical cancer is at the forth position of the most diagnosed cancer and death about cancer.It is essential to distinguish the progression of different patients with precancerosis or carcinoma timely,because of development of cervical cancer.PAX-1 gene as a regulatory gene,with abnormal methylation shows a close relation with cervical cancer.Method and material:52 patients` cervical specimens from The First Affiliated Hospital of Chongqing Medical University were collected.All patients have TCT test,colposcope test and necessary pathological examination.The cervical samples were collected with TCT test,and DNA purification was carrying on molecule lab.PCR amplification technique was used to experience the level of PAX-1 methylation.Using statistics methods to analyse data.Consequence: PAX-1 methylation have a high expression among HSIL and cancer tissue,Total negative predictive value is 51.6% and positive predictive value gets to 100%;there are 6hypermethylated tissues among HPV 16/18 patients with HSIL and Cancer at 27.3%;HPV-Test combined PAX1 will improve diagnose value to63%.Conclusion:with close relationship between PAX-1 methylation andHSIL and cancer tissue,the test of PAX-1 methyaltion is helpful to diagnosis patient with cervical lesion especially HSIL,and find possibility of cancer.As same time PAX1 methylation can improve the value of HPV test.
