| Objective: To detect expression level of LDOC1 in colorectal cancer,and to investigate its biological function and related molecular mechanism in colorectal cancer.It provides a new target for early diagnosis and effective treatment of colorectal cancer.Methods: The expression of LDOC1 in colorectal cancer tissues and adjacentnormal tissues was detected by quantitative real-time PCR(qRT-PCR)and immunohistochemistry(IHC).LDOC1 expression in four CRC cell lines compared with normal colorectal tissue was determined by reverse transcription-polymerase chain reaction(RT-PCR),and two cell lines with relatively low expression were screened.LDOC1 overexpressed lentivirus and emptylentivirus weretransfected into HCT-116 and Caco2 cell lines and the transfection efficiency was identified by RT-PCR and western-blot.Cell proliferation was detected by CCK-8 and colony formation assays.Cell cycle and apoptosis were detected by flow cytometry assay.Migration and invasion were assessed using transwell assay.In addition,LDOC1 regulates Wnt/?-catenin pathway and epithelial-mesenchymal transition(EMT)of colorectal cancer cells were investigated by western-blot,and the expression and localization of?-catenin in colorectal cancer cells were demonstrated by cellular immunofluorescence.Results: LDOC1 expression was down-regulated in colorectal cancer tissues and cells.LDOC1 overexpression inhibited cell proliferation,migration,invasion,cell cycle arrest but promoted cells apoptosis.At the same time,after overexpression of LDOC1,pro-apoptotic proteins increased significantly and anti-apoptotic proteins decreased significantly.Furthermore,LDOC1 inhibits the metastasis and EMT of colorectal cancer cells by down-regulating the Wnt/ ?-catenin signaling pathway.Conclusion: LDOC1 is a tumour suppressor in CRC and it inhibits cell proliferation and promotes cell apoptosis.In addition,it inhibits colorectal cancer cell metastasis by down-regulating the Wnt/?-catenin signaling pathway. |
