Objective:The aim of this study was to investigate the preventive and therapeutic effects of ErzhiPillson1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-inducedParkinson disease in mice.Methods:Thirty healthy and active mice were randomly divided into the controlled group,the model group and the drug-administered group,with 10 mice in each group.The mice in the drug-administered group were pre-administered with Erzhi Pills 2.5 g·kg-1,0.1ml·10g-11 for 10 days.The controlled group and the model group were given equal volume of normal saline.From the 11th to 18th days,both the model group and the drug-administered group were intraperitoneally injected with MPTP,30 mg·kg-1,0.1ml·10g-11 once daily to induce PD,while the mice of the controlled group were given equal volume of normal saline.Then the mice of drug-administered group was intragastrically administered with Erzhi Pills,and the controlled group and the model group were given the same volume of normal saline from the 19th to 25th days.From the 26th to 28th days,the Morris water maze positioning navigation experiment was carried out,and the space exploration experiment was carried out on the 30th day.During the period of administration,the mice of drug-administered group were continuously administered with two pills until the end of the experiment.In the behavioral aspect,the limb movement coordination ability of the mice was detected by climbing rod experiment,hanging experiment and swimming experiment.Morris water maze was used to detect the spatial learning and memory ability of the mice.The content of MDA,the activity of GSH-PX and SOD in serum were determined by spectrophotometry.Western Blot was used to detect the expression levels of TH,MAO-B and apoptosis-related factors CHOP and Caspase-12 in brain tissue.To observe the damage of dopaminergic nerve in the substantia nigra,immunohistochemistry was used to observe the number of TH-positive cells in the brain tissue of mice.Results:In terms of behavioral testing,compared with the controlled group,the limb movement scores and the ability of spatial learning and memory of the model group mice were significantly lower?P<0.05,P<0.01?,while compared with the model group mice,the scores of limb movement and the ability of spatial learning and memory of the drug-administered group mice were significantly increased?P<0.05,P<0.01?.Compared with the controlled group mice,the activities of SOD and GSH-PXin the serum of the model group were significantly reduced,and the content of MDA was significantly increased?P<0.05,P<0.01?;compared with the model group mice,the activities of SOD and GSH-PX in the serum of the drug-administered group mice were significantly increased?P<0.05,P<0.01?,and the content of MDA was significantly diminished?P<0.05,P<0.01?.The results of Western Blot showed that compared with the controlled group,the protein levels of TH in the brain tissue of the mice of the model group were significantly decreased?P<0.05,P<0.01?,and the protein levels of MAO-B and apoptosis-related factors CHOP and Caspase-12 were significantly up-regulated?P<0.05,P<0.01?;compared with the model group,the protein level of TH in brain tissue of the drug-administered group was significantly increased,and the protein levels of MAO-B and apoptosis-related factors CHOP and Caspase-12 were significantly down-regulated?P<0.05,P<0.01?.The results of immunohistochemistry showed that compared with the controlled group,the number of TH-positive cells in the model group was significantly reduced?P<0.05,P<0.01?,while compared with the model group,the group of drug-administered was contrary?P<0.05,P<0.01?.Conclusion:These studies indicate that Erzhi Pills has a certain preventive effect on MPTP-induced PD mice.Erzhi Pill can significantly improve the coordination of limb movement and spatial learning and memory in PD mice,which may be related to inhibition of oxidative stress,down-regulation of apoptosis-related factors CHOP,Caspase-12 expression,reduction of dopaminergic neuron damage,inhibition of dopaminergic neuronal apoptosis.
