Objective: To investigate the expression of RCAN 1 transcript in esophageal squamous cell carcinoma and its correlation with clinicopathology and prognosis.Methods: 103 specimens of esophageal squamous cell carcinoma patients and their matched paracancerous tissues were selected from the Fourth Affiliated Hospital of Henan University of Science and Technology(Anyang Cancer Hospital)from February 2014 to September 2017.They were used as experimental group(TT)and control group(PT),and the corresponding clinical and pathological data were collected.TT and PT tissues were taken to extract RNA,QRT-PCR was used to detect the expression of RCAN 1 in cancer and adjacent tissues.SPSS 17.0 software was used to analyze the data.The correlation between RCAN 1 and esophageal squamous cell carcinoma was explored,and the correlation between the expression of RCAN 1 and clinical pathological development and prognosis of patients was analyzed.Results: The expression levels of RCAN 1.1,RCAN 1.2 and RCAN 1.4 transcripts in esophageal squamous cell carcinoma tissues were lower than those in adjacent tissues(p < 0.001,p = 0.031,p = 0.055).The expression levels of RCAN 1.1 and RCAN 1.2 were significantly different.The cutoff values of RCAN 1.1,RCAN 1.2 and RCAN 1.4 transcripts were obtained from ROC curves.The subjects whose expression of RCAN 1 was lower than the cutoff value in the TT group were classified into low expression group and those whose expression was higher than cutoff value were classified into high expression group.The relationship between them and clinicopathological features was analyzed by Chi-square test.1.The correlation analysis of RCAN 1.1 expression level and clinicopathological characteristics were as follows: RCAN 1.1 expression was correlated with TNM stage,and low expression of RCAN 1.1 was more common in stage II and III esophageal squamous cell carcinomas(p = 0.013);RCAN 1.1 expression was correlated with depth of invasion,with statistical significance(p = 0.048).The expression of RCAN 1.1 was not correlated with the degree of differentiation,the location of the disease and lymph node metastasis.2.The correlation analysis between RCAN 1.2 expression level and clinicopathological features was as follows: RCAN 1.2 expression level was correlated with TNM stage,RCAN 1.2 was highly expressed in stage I esophageal squamous cell carcinoma,and low expressed in stage II and III esophageal cancer,with statistical significance(p < 0.001);RCAN 1.2 expression level was significantly correlated with depth of invasion(p < 0.001),invasion(p < 0.001).The expression of RCAN 1.2 was not related to the degree of differentiation,location of disease and lymph node metastasis.The expression of RCAN 1.2 was correlated with the total survival time(p = 0.001).3.The correlation analysis of RCAN 1.4 expression level and Clinicopathology and prognosis showed that there was statistical significance between RCAN 1.4 and the degree of differentiation of esophageal squamous cell carcinoma(p < 0.001),the distribution of high expression in differentiated esophageal squamous cell carcinoma and low expression in poorly differentiated tumors.RCAN 1.4 was not related to TNM stage,site of onset,depth of invasion and lymph node metastasis.Kaplan-Meier univariate survival and prognosis analysis showed that RCAN 1.2,location of onset,TNM stage,T stage,and N stage were significantly different from the total survival time(OS)of ESCC patients(p = 0.001,p = 0.039,p < 0.001,p < 0.001,p < 0.001,p < 0.001).Multivariate analysis showed that TNM stage,T stage,N stage,site of onset and RCAN 1.2 were not independent prognostic factors for ESCC.Tumor differentiation was independent prognostic factor(p = 0.037,95% CI:0.806-5.519).Conclusion: The expression of RCAN I.1,RCAN 1.2 and RCAN 1.4 transcripts in esophageal squamous cell carcinomas were decreased,and the expression of RCAN 1.1 and RCAN 1.2 had a significant difference.The expression levels of RCAN I.1 and RCAN 1.2 were negatively correlated with TNM staging and invasion depth of esophageal squamous cell carcinoma,while RCAN 1.4 was negatively correlated with the differentiation degree of esophageal squamous cell carcinoma.RCAN 1.2 is associated with survival of esophageal squamous cell carcinoma.Patients with high RCAN 1.2 expression have shorter overall survival than those with low RCAN 1.2 expression.
