| Objective:In recent years,the incidence of skin tumors has been increasing year by year,which has increased the economic burden for people,especially for some malignant tumors,which are easy to be ignored in the early stage of disease and have seriously threatened life when they go to see a doctor.Therefore,it is of far-reaching significance to look for indicators related to benign and malignant skin tumors and make a hint for the prognosis of the disease.In this study,MMP13,KGF,IL-6 and IGFBP3 will be tested in SCC,BD and SK patients,and their significance will be analyzed,so as to provide reference for the prognosis of the disease.Methods:1.The pathological tissues of skin squamous cell carcinoma,Bowen's disease,seborrheic keratosis and normal tissues around pigmented nevus that were surgically divided from the affiliated hospital of Chengde medical college and Cangzhou central hospital were collected.According to the disease type,the tissues were divided into 4 groups,namely skin squamous cell carcinoma group,Bowen's disease group,seborrheic keratosis group and normal control group,SP immunohistochemical stain was used to detect the expression of MMP13,KGF,IL-6,IGFBP3 in 26 cases of skin squamous cell carcinoma,21 cases of Bowen's disease,48 seborrheic keratosis and 10 normal skin tissues.2.SPSS19.0 software was used for statistical analysis,the counting data were represented by n(%),the chi-square test was used for the comparison between groups,the measurement data were represented by X ±s,the difference was statistically significant at P < 0.05,Bonferroni method was used for the multiple comparisons of rates,differences were considered significant at P < 0.0083.Results:1.The positive rates of MMP13 in skin lesions of patients in SCC group,BD group,SK group and normal tissue group were 80.77%(21/26),90.48%(19/21),37.5%(18/48)and 0(0/10),respectively.The pairwise comparison shows that there was no significant difference between SCC group and BD group(?2=0.268,P=0.605),there was significant difference between SCC group and SK group(?2=12.667,P=0.000),there was significant difference between SCC group and normal group(P=0.000),there was significant difference between BD group and SK group(?2=14.425,P=0.000),there was significant difference between BD group and normal group(P=0.000),and there was significant difference between SK group and normal group(P=0.000).2.The positive rates of KGF in SCC group,BD group,SK group and normal tissue group were 80.77%(21/26),42.86%(9/21),27.08%(13/48)and 0(0/10),respectively.The pairwise comparison shows that there was significant difference between SCC group and BD group(?2=7.232,P=0.007),there was significant difference between SCC group and SK group(?2=19.572,P=0.000),there was significant difference between SCC group and normal group(P=0.000).There was no significant difference between BD group and SK group(?2=1.674,P=0.196),there was no significant difference between BD group and normal group(P=0.030),and there was significant difference between SK group and normal group(P=0.000).3.The positive rates of IL-6 in SCC group,BD group,SK group and normal tissue group were 57.69%(15/26),47.62%(10/21),4.17%(2/48)and 0(0/10),respectively.The pairwise comparison shows that there was no significant difference between SCC group and BD group(?2=0.473,P=0.491),there was significant difference between SCC group and SK group(?2=24.364,P=0.000),there was significant difference between SCC group and SK group(P=0.002).There was statistical difference in BD group and SK group(?2=16.294,P=0.000),there was no significant difference between BD group and normal group(P=0.012),and there was no significant difference between SK group and normal group(P=1.000).4.The positive rates of IGFBP3 in skin lesions of patients in SCC group,BD group,SK group and normal tissue group were 38.46%(10/26),52.38%(11/21),93.75%(45/48)and 100%(10/10),respectively.The pairwise comparison shows that there was no significant difference between SCC group and BD group(?2=0.911,P=0.340),there was significant difference between SCC group and SK group(?2=24.195,P=0.000),there was significant difference between SCC group and SK group(P=0.001).There was statistical difference in BD group and SK group(?2=13.757,P=0.000),there was no significant difference in BD group and normal group(P=0.012),and there was no significant difference in SK group and normal group(P=1.000).Conclusion:1.The differences of MMP13 between SCC group and SK group,normal group and BD group and SK group,normal group and SK group were statistically significant,indicating that MMP13 may play a role in the progression of sebaceous keratosis,squamous cell carcinoma in situ and skin infiltrating squamous cell carcinoma,but the differences of MMP13 between SCC group and BD group were not statistically significant,which suggests that MMP13 may have no predictive and referential significance for the malignant degree of skin squamous cell carcinoma.2.There was significant difference between SCC group and BD group,SCC group and SK group,SCC group and normal group,and BD group and SK group,indicating that KGF may provide a reference for the progress and prognosis of seborrheic keratosis,squamous cell carcinoma in situ and skin infiltrating squamous cell carcinoma.Meanwhile,there was no significant difference between SK group and normal group,indicating that KGF may not play a role in promoting the occurrence of seborrheic keratosis.However,there was no significant difference between the BD group and the normal group,which may be related to the small sample capacity.3.There was significant difference of IL-6 between SCC group and SK group,SCC group and normal group,BD group and SK group,and SK group and normal group,but there was no significant difference between SCC group and BD group,suggesting that IL-6 may play a role in the occurrence and development of seborrheic keratosis,and skin squamous cell carcinoma.However,it has poor prognostic effect in predicting whether squamous cell carcinoma has metastasis.However,there was no significant difference between the BD group and the normal group,which may be related to the small sample capacity.4.There was significant difference of IGFBP3 between SCC group and SK group,SCC group and normal group,BD group and SK group,indicating that IGFBP3 may provide a reference for the differentiation of cutaneous squamous cell carcinoma and sebaceous keratosis,but there was no significant difference between SCC group and BD group,indicating that MMP13 may not be able to indicate whether cutaneous squamous cell carcinoma has metastasis.There was no significant difference between SK group and normal group,indicating that IGFBP3 may not play a role in promoting the occurrence of seborrheic keratosis.However,there was no statistical significance between the BD group and the normal group,which may be related to the small sample capacity. |
