ObjectiveBy detecting and observing the expression of MIF and GSK-3? in laryngeal squamous cell carcinoma,the correlation between the expressio n of MIF and GSK-3? and the clinicopathological factors of laryngeal squamous cell carcinoma was analyzed,and the occurrence and developm ent of laryngeal carcinoma,and the relationship between the two were explored.To provide a new theoretical basis for the clinical diagnosis and treatment of laryngeal squamous cell carcinoma,and detect the disease progression in patients with laryngeal cancer.Provide a reference for prognostic evaluation.Methods1.From April 2017 to July 2018,a collection of tumor tissue specimens from 30 patients with primary laryngeal squamous cell carcinom a in the Department of Otorhinolaryngology,Chaoyang Central Hospital,Liaoning Province,all specimens have corresponding adjacent paracancer ous tissues(at least 0.5cm from the margin of the tumor),and the normal mucosal tissues of the throat of 10 patients with non-larynx cancer were compared.2.All specimens were fixed in formalin and embedded in paraffin.Immunohistochemical staining was used to detect the expression of MIF and GSK-3? in laryngeal squamous cell carcinoma,paracancerous tissues and normal laryngeal mucosa.3.Collect the clinical data of patients,analyze the relationship bet ween the expression of MIF and GSK-3? in laryngeal squamous cell carcinoma and clinicopathological factors,and explore the correlation between MIF and GSK-3? expression in laryngeal squamous cell carcinoma.Results1.MIF is expressed in laryngeal squamous cell carcinoma,adjacent tissues and normal mucosa of the larynx.The expression of MIF in laryngeal squamous cell carcinoma(positive rate 73.33%)is higher than that in adjacent tissues(positive rate 40.00%)and the normal mucosa(positive rate 20.00%),the difference was statistically significant(p<0.05);there was no significant difference in the expression of MIF between adj acent tissues and normal mucosa,p>0.05.2.There was a significant difference in MIF between laryngeal squamous cell carcinoma and different pathological differentiation groups(p=0.036).The expression of MIF in the poorly differentiated group was higher than that in the moderately and highly differentiated group,the ex pression in the moderately differentiated group was higher than that in the highly differentiated group;The expression of MIF in the clinical stage III-IV was significantly higher than that in the clinical stage I-II group,the difference was statistically significant(p=0.049);the expression of MIF in the lymph node metastasis group was higher than that in the no lymph node metastasis group(p=0.029);There was no significant dif ference in the expression of MIF between age,gender,smoking history and drinking history(p>0.05).3.GSK-3? is expressed in laryngeal squamous cell carcinoma,adjacent tissues and normal mucosa of the larynx.The expression of GSK-3? in laryngeal squamous cell carcinoma(positive rate 70.00%)is higher than that in adjacent tissues(positive rate 13.33%)and normal larynge almucosa(positive rate 20.00%),the difference was statistically significa nt(p<0.05);there was no significant difference in the expression of GSK-3? between adjacent tissues and normal mucosa,p>0.05.4.GSK-3? was significantly different in laryngeal squamous cell carcinoma tissues with different pathological differentiation(p=0.011).The expression of GSK-3? in the poorly differentiated group was higher tha n that in the moderately and highly differentiated group,the expression in the moderately differentiated group was higher than that in the highly differentiated group;The expression of GSK-3? in the clinical stage III IV was significantly higher than that in the clinical stage I-II group,the difference was statistically significant(p=0.020);the expression of GSK-3? in the lymph node metastasis group was higher than that in the no lymph node metastasis group,with significant difference(p=0.013);there was no significant difference in the expression of GSK-3? between age,sex,smoking history and drinking history(p>0.05).5.In laryngeal carcinoma,the expression of MIF and GSK-3? was positively correlated,and the correlation coefficient was r=0.592.The co rrelation was statistically significant(p=0.003).Conclusion1.The high expression of MIF and GSK-3? in laryngeal squamous cell carcinoma suggests that MIF and GSK-3? may play a role in promoting oncogene proliferation in laryngeal squamous cell carcinoma,sugge sting that MIF and GSK-3? may be potential for diagnosis of laryngeal cancer molecular markers.2.The expression of MIF and GSK-3? in laryngeal squamous cell carcinoma was correlated with clinical stage,pathological differentiation and lymph node metastasis,suggesting that the high expression of MIF and GSK-3? is related to the invasion and metastasis of laryngeal squamous cell carcinoma.3.The expression of MIF and GSK-3? in laryngeal squamous cell carcinoma was positively correlated,suggesting that MIF may positively regulate the expression of GSK-3?.4.MIF and GSK-3? can be used as potential molecular biomarkers for laryngeal cancer.They can be used as a reference for diagnosis and treatment of laryngeal cancer,monitoring of disease progression and prognosis.They provide a theory for reversing the development of laryngeal cancer and the application of gene-targeted drugs reference.
