Salvia miltiorrhiza Bge.and Carthamus tinctorius L.are two kind of traditional Chinese herbal medicines,which has a long history of compatibility.Modern pharmacological studies have found that salvia miltiorrhiza and safflower have a kind of pharmacologically active components.Danhong injection has a good effect on cardiovascular diseases,which has been on the market for many years.This study was designed to investigate the protective effects of active ingredients in Danhong on cognitive dysfunction and its mechanisms,which is caused by hypoperfusion of whole brain.And provide relevant experimental basis for its clinical application.In this thesis,salvia miltiorrhiza Bge.and safflower yellow A were obtained by acid extraction,macroporous resin chromatography,reflux and extraction.The whole brain hypoperfusion model of mice was constructed by surgery to explore the protective effect and mechanism of active ingredients in Danhong on cognitive dysfunction caused by hypoperfusion.The main research contents and results are as follows:(1)The preparation method is simple and feasible,the cost is low,the operation is safe,and the final product obtained has a purity of 98% and less impurities.(2)A mouse model of whole brain hypoperfusion was established by surgically placing a coil in the common carotid artery of the mouse to cause common carotid artery stenosis.The modeling method is simple and feasible,and the survival rate can reach 75%~80%.The success of the model was verified by a method of testing toluidine blue O staining by Morris water maze.The percentage of total time in the escape latency,target quadrant stay time,the percentage of total distance from the target quadrant,and the number of crossing platforms were significantly different between the mice one month after surgery and the normal group.In the experiment of toluidine blue O staining,there were also significant differences between the model group mice and the normal group mice.(3)The laboratory extract of salvianolic acid B and hydroxysafflor yellow A was used to study the protective effect of cerebral acetylcholinesterase induced by whole brain hypoperfusion.The content of acetylcholinesterase was determined by Western blot.The phosphorylation level of Ser-262 was detected by Western blot,and its protective mechanism was initially explored.The experiment found that the active ingredients in Danhong can alleviate the cognitive dysfunction caused by hypoperfusion in the whole brain and improve the ability of learning and memory.The active ingredient in Danhong can reduce the activity of acetylcholinesterase in the brain.Salvianolic acid B,hydroxysafflor yellow A and the combination of the two can significantly reduce the phosphorylation level of Tau protein Ser-262.
