BACKGROUND: Renal cancer is one of the most common malignancies of the urinary system and accounts for approximately 2-3% of all adult malignancies.Clear cell renal cell carcinoma is the most common type of renal cell carcinoma,accounting for approximately 85% of all renal cell carcinomas.Many studies have shown that the development of clear cell renal cell carcinoma is regulated by various molecules.Renalase is a protease that is mainly synthesized and secreted by renal tubular epithelial cells and can regulate blood pressure by breaking down catecholamines and plays an important role in human diseases.In recent years,significant advances have been made in the study of the structure,function,action mechanism and relationship with renalase.Renalase has been found to have differential expression in normal tissues and tumor tissues,such as pancreatic cancer and bladder cancer.Renalase is highly expressed in breast cancer,melanoma,and the like.Renalase may be involved in the process of tumorigenesis and development.Related studies have found that abnormally elevated expression of renalase is beneficial to the survival of cancer cells and promotes tumor growth.The inhibition of renalase signaling may become a new therapeutic option for cancer.At present,there is no clinically relevant study on the expression of renalase in renal tissue of patients with clear cell renal cell carcinoma.Objective: The study aims to investigate the expression of renalase in the renal tissues and normal kidney tissues of patients with clear cell renal cell carcinoma and to investigate the renalase expression levels in renal tissues of patients with clear cell renal cell carcinoma and non-cancer patients.It analyzes the expression levels of renalase in renal tissues of patients with clear cell renal cell carcinoma whether is correlated with tumor size(maximum tumor size),clinical stage,uric acid,serum sodium and age.Its application values in clinical diagnosis and its roles in the development of clear cell renal cell carcinoma are speculated.And renalse may provide a new direction for the diagnosis and treatment of renal cancer.METHODS: The patients with clear cell renal cell carcinoma and renal contusion who had undergone surgical treatment in the Department of Urology,Zhongda Hospital from July 2015 to July 2017 are selected.Patient tissue waxes are obtained from Department of Pathology,Zhongda Hospital(including cancerous tissue and normal kidney tissue).All the patients are tested for relevant laboratory indicators before surgery,and the tumor size(maximum tumor size)and clinical stage were measured after surgery.The pathological tissues are made into tissue sections and immunostained with anti-renalase antibody.The results of immunohistochemical staining are analyzed semi-quantitatively using Image-Pro Plus 6.0 software.The expressions of renalase in renal tissues are compared by comparing the iod(sum)values of different samples in the same area.The relevant data is analyzed using statistical software SPSS 19.0.Chi-square test,t test,Pearson correlation analysis,Spearman correlation analysis,and multi-factor regression analysis.Results: 1.Renalase is negatively expressed in renal cell carcinoma in patients with clear cell renal cell carcinoma and is positively expressed in normal kidney tissue.Renalase is mainly expressed in the cytoplasm of renal tubular epithelial cells.The positive expressions of renalase is brown-yellow and brown-brown.2.There is a significant difference in age,serum sodium,hemoglobin and uric acid between the clear cell renal cell carcinoma group and the renal contusion group.3.The iod values of clear cell renal cell carcinoma group are significantly higher than that of renal contusion group(P<0.05).The expression levels of renalse in normal kidney tissue of patients with clear cell renal cell carcinoma are significantly higher than that of patients with renal contusion.4.The expression levels of renalse in renal tissue of patients with clear cell renal cell carcinoma are positively correlated with tumor size(maximum tumor size)and clinical stage,but not with uric acid,serum sodium,and age.Conclusion: 1.Renalase is negatively expressed in renal cell carcinoma of clear cell renal cell carcinoma and positively expressed in normal kidney tissue.2.The expression levels of renalase in renal tissue of patients with clear cell renal cell carcinoma are significantly higher than that of non-cancer patients.3.The expression levels of renalase in renal tissue of patients with clear cell renal cell carcinoma are positively correlated with the tumor size(maximum tumor diameter)and clinical stage of renal cancer patients.
