Background and ObjectiveGpc3(Glypican-3)modulates cellular proliferation,differentiation and genic expression structurally and functionally.Recent researches find that Gpc3 has a quite important role in calvarial osteogenesis and fusion of cranial sutures.This study aimed on exploring Gpc3 expression in cranial suture tissues and cells of different stages,and providing reference for later research in disease models.MethodsStudy and observe cranial suture tissues and cells of 1d,3d,7d in SD(Sprague Dawley)rats.Analyze expression level of Gpc3 with immunohistofluorescence staining,RT-qPCR(Reverse transcription quantitative real-time polymerase chain reaction)and western blotting.And judge whether there is a relationship with Gpc3 expression and calvarial osteogenesis/fusion of cranial sutures by the result of calvarial micro CT(Computed Tomography)scan.ResultsGPC3 protein was expressed in all stages of unfused rat cranial suture tissues.Result of RT-qPCR showed a descending trend with the age increasing.Result of western blotting indicated that GPC3 protein was detected in all groups of cells.Result of calvarial micro CT scan showed that postfrontal suture was fused on 7th day postnatal,while the sagittal and coronal sutures were fused on the 14 th day postnatal.Conclusions1)Gpc3 is expressed in all stages of unfused rat cranial suture tissues and the expression level descends with the fusing of cranial sutures.2)GPC3 protein is secreted to extracellular matrix and may have an important role in modulation of calvarial osteogenesis and fusion of cranial sutures.
