| Cytokine-induced killer(CIK)cells were endowed with the characteristic of high proliferative capacity,high cytotoxicity,broad anti-tumor spectrum and targeted tumor therapy.Gold nanoprisms(AuNPrs)have drawn much attention due to their unique optical properties and the potential applications such as photoacoustic imaging and photothermal therapy.This dissertation focus on the preparation of multifunction probe,CIK-PEGylated AuNPrs,owned the characteristics of both CIK cells and AuNPrs,which exhibit great potential in applications such as targeted photoacoustic imaging and photothermal therapy of gastric cancer in the future.The main results of this study are as follows:1.To explore the effect of cytokine induced killer(CIK)cells on proliferation,migration and the cycle of MGC803 cells,we incubated the CIK cells with MGC803 cells on different incubation time and E:T ratios.The results showed that CIK cells could markedly influence the proliferation and migration of MGC803 cells and inhibited the cell cycle in G1 phase;CIK cells stained with DiR were injected to mice to observe the distribution of CIK cells by animal fluorescence imaging system.Fluorescence imaging in vivo showed that CIK cells had the characteristic to target the gastric cancer site.The experimental results indicate that CIK cells can targeted kill gastric cancer in vivo.2.The gold nanoprisms(AuNPrs)here were synthesized by reducing HAuCl4·3H2O with Na2S2O3,a straightforward method without any using of toxic reagents.The AuNPrs have three congruent edge lengths of 100nm and uniform thickness of 10nm,and display a surface plasmon resonance(SPR)band in the near infrared area,which is a suitable wavelength for photoacoustic imaging and photothermal therapy purposes.Experiments in vitro demostrate the reducing cytotoxicity and high light-thermal conversion efficiency of AuNPrs.Illuminate the MGC803cells incubated with AuNPrs,the viability of MGC803 decreased significantly.3.On the basis of the work above,CIK cells labeled with PEGylated AuNPrs were successfully prepared,demonstrated by dark field microscope and two-photon microscope.Owned the characteristics of CIK cells and AuNPrs,the probe was successfully used for targeted photoacoustic imaging,enhanced immunotherapy and photothermal therapy of Gastric Cancer,providing a new way for gastric cancer therapy in future. |
