OBJECTIVE:To explore the regulation of electroacupuncture and anti-depression central mechanism by electroacupuncture based on Wnt/ ?-catenin signal transduction pathway in the behavioral and pathwayal expression of key proteins and hippocampal neuroremodeling in rats with depression.METHODS:144 male Sprague-Dawley rats were randomly divided into 4 groups,including blank,model,electroacupuncture and fluoxetine group.Each group was further divided into 3 subgroups according to different intervention time: 7d,14 d and 21 d..The blank group of animals did not receive any model stimulation and were fed freely.Rats in the model group were constructed with a CUMS depression model by fasting or no water and etc.The electroacupuncture group selected Baihui and Shenting points,and the electroacupuncture was applied once a day.The fluoxetine group was given fluoxetine once a day after daily stress stimulation.The changes of behavioral indexes of the above groups were observed on the 7th,14 th and 21 st,respectively.Western-Blot was used to detect the key proteins Wnt1,?-catenin and GSK-3 ? in the Wnt/?-catenin signal transduction pathway in the hippocampus of rats.The morphological changes of neurons in hippocampal CA3 area were observed by transmission electron microscopy.Data statistics were analyzed by SPSS22.0 software.RESULT:1.Behavioral changes in rats with depressionBefore the construction of the successful model,the differences in behavioral changes such as the open field test score,sucrose water preference difference and body mass evaluation of the experimental groups were not statistically significant(p>0.05).On the 7th day of the experiment,compared with the blank group,the three behavioral indicators of the model group were decreased,and the difference was statistically significant(p<0.01).Compared with the model group,the behavior of the electroacupuncture and fluoxetine group rats Except for body mass evaluation,the three indicators of learning were improved to some extent,but the difference was not statistically significant(p>0.05).On the 14 th and21st day of the experiment,compared with the blank group,the three behavioral indexes of the model group were significantly lower,and the difference was statistically significant(p<0.01).Compared with the model group,the behavior of the electroacupuncture and fluoxetine group rats All three indicators of learning were up,and the difference was statistically significant(p<0.05).There were no significant differences in the behavioral parameters between the western medicine and the electroacupuncture group(p>0.05).2.The regulation of electroacupuncture on the expression of key proteins in Wnt/?-catenin signaling pathwayOn the 7th day,compared with the blank group,the expression levels of Wnt1 and ?-catenin in the brain of the model group decreased slightly,and the expression level of GSK-3 ? protein increased.The difference was statistically significant(all p<0.05).Compared with the model group,the expression levels of Wnt1 and ?-catenin in the brain of rats in the electroacupuncture and fluoxetine group increased slightly,and the expression level of GSK-3? protein decreased,and the difference was statistically significant(p<0.05).On the 14 th and 21 st day of experiment,compared with the blank group,the expression levels of Wnt1 and ?-catenin in the brain of the model group decreased,and the expression level of GSK-3? protein increased.The difference was statistically significant(p<0.05).Compared with the model group,the expression of Wnt1 and ?-catenin protein in the brain of electroacupuncture and western medicine intervention group increased,and the expression level of GSK-3? protein decreased,the difference was statistically significant(p<0.05)..Moreover,the expression levels of each group in the brain of electroacupuncture and fluoxetine group were compared between the groups during the intervention period,and the difference was not statistically significant(p>0.05).3.Morphological and synaptic plasticity changes in rat hippocampal neuronsOn the 7th day of experiment,the neurons in the hippocampal CA3 area of ??the blank group were densely ordered,and the synaptic structure was firm and firm,and the shape was full and full.Compared with the neuronal structure and morphological changes in the hippocampal CA3 area of the rats in the blank group,the neurons in the hippocampal CA3 area of the model group decreased,the number of synapses decreased,the apoptotic bodies increased,and the cell arrangement gradually became scattered and vacuolated.Morphological shrinkage,structural framework dissolution,gradual deformation,and synaptic number group comparison,the difference was statistically significant(p <0.01).Onthe 14 th and 21 st day of the experiment,compared with the blank group,the hippocampal CA3 area of the model group decreased,and the number of synapses decreased,the shape was shrunk,the structural framework was dissolved and deformed,and the number of synapses was compared.The difference was statistically significant(p<0.01).Compared with the model group,the neurons in the hippocampal CA3 area of the electroacupuncture and fluoxetine groups increased,the number of synapses increased,the morphology became full and full,and the structural framework was relatively complete.The difference between the groups of synaptic numbers was statistically significant(p<0.05).There was no significant difference in the number of synapses between the electroacupuncture and the fluoxetine group during the intervention period(p>0.05).CONCLUSION:1.Electroacupuncture is effective in improving the behavioral characteristics of depressed rats and alleviating depressive symptoms.2.Electroacupuncture can alleviate the symptoms of depression by regulating the expression of key proteins in the Wnt/?-catenin signal transduction pathway in hippocampus,which may be one of the central mechanisms of electroacupuncture antidepressant central effect.3.Electroacupuncture can be used to regulate the morphology and synaptic plasticity of hippocampal neurons,effectively inhibit neuronal damage and degeneration,indirectly protect nerves and induce nerve remodeling.
