Objective:Research the effect of Yigan Capsule on TGF-?1/STAT3 signal pathway in mice with immunological liver injury induced by ConA.The aim is to provide a theoretical basis for exploring a new target for clinical treatment of immunological liver injury.Methods:50 SPF mice were taken and divided into 5 groups randomly:model group,normal group,high dose group,medium dose group and low dose group of Yigan capsule,given gavage for 2 weeks(The dosage was 1.872g/kg in high dose group,0.936g/kg in medium dose group and 0.468g/kg in low dose group).1 hour after the last gavage,mice were injected with 20mg/kg ConA to establish a model of immunological liver injury,and the mice were killed after 8 hours.HE staining was used to observe the pathological changes of liver tissue microscopically.ALT and AST in serum of mice were detected by biochemical method.The levels of TGF-?1,ICAM-1,IL-1? and IL-6 in serum were detected by ELISA.The expression of TGF-?1 and ICAM-1 in liver tissue was detected by immunohistochemistry.The expression of p-JAK2 and p-STAT3 proteins was detected by western blotting.Results:1.In the model group,the liver tissue structure was disordered,with a large number of inflammatory cells infiltrating,local liver tissue swelling,fatty degeneration or necrosis.The pathological damage of each dose group of Yigan capsule was alleviated to different degrees,the number of inflammatory cells was significantly reduced,the structure of liver cells was relatively regular,local liver tissue edema was alleviated,and slight fatty degeneration was observed,especially in the high dose group.2.Compared with the normal group,the serum levels of ALT,AST,TGF-?1,ICAM-1,IL-1? and IL-6 in the model group were significantly increased(P<0.01).Compared with the model group,the above-mentioned indexes in each dose group of Yigan Capsule decreased in different degrees(P<0.01),especially in the high dose group.3.Compared with the normal group,the expression of TGF-?1,ICAM-1,p-JAK2 and p-STAT3 in the liver tissue of the model group was significantly increased(P<0.01).Compared with the model group,the expression of TGF-?1,ICAM-1,p-JAK2 and p-STAT3 in each dose group of Yigan capsule decreased in different degrees(P<0.01),especially in the high dose group.Conclusion:1.Yigan Capsule can significantly improve the liver function and alleviate the pathological damage of liver tissue of mice with immunological liver injury induced by ConA.2.Yigan Capsule has obvious inhibitory effect on inflammatory cytokines TGF-?1,ICAM-1,IL-1? and IL-6 in serum of mice with immunological liver injury induced by ConA.3.Yigan Capsule can significantly inhibit the expression of TGF-?1,ICAM-1,p-JAK2 and p-STAT3 in liver tissue of mice with immunological liver injury induced by ConA.In conclusion,Yigan Capsule may play a regulatory role in TGF-?1/STAT3 signaling pathway by regulating the phosphorylation level of JAK2 and inhibiting inflammatory cascade reaction,in order to improve the liver function and pathological damage of mice with immunological liver injury induced by ConA.
