Objective:In recent years,there are a few literatures about the relationship between TRIM35 and non-small cell lung cancer.The purpose of this study is to investigate the expression of TRIM35 in non-small cell lung cancer,para-cancerous tissues and normal lung tissues,and to explore the clinical and pathological characteristics of patients with non-small cell lung cancer.And the relationship between TRIM35 and survival and recurrence of patients with non-small cell lung cancer will be expounded.The significance of this paper is to provide experimental basis for further studies on the correlation between TRIM35 and non-small cell lung cancer.Methods:1.The expression of TRIM 35 protein in the tumor tissue,40 adjacent tissues and 40 normal lung tissues of 148 patients with non-small cell lung cancer were measured by tissue microarray and immunohistochemistry.The148 patients were all treated with radical surgery in the department of thoracic surgery of the fourth hospital of Hebei medical university from August 2006to January 2014.2.The expression of TRIM35 in various tissues was detected by immunohistochemistry.3.The clinical data of patients included age of onset,smoking history,drinking history,family history,lymph node metastasis,distant metastasis,clinical stage and clinicopathological features of tumors.4.The relationship between TRIM35 expression and clinicopathological features of NSCLC was analyzed by chi-square test and rank correlation analysis.Kaplan-Meier analysis was used for survival analysis.Cox regression multivariate analysis was used to analyze the effect of TRIM35 expression on the prognosis of patients with non-small cell lung cancer after operation.All statistical analysis was carried out by SPSS25.0 statistical software package.Results:The expression of TRIM35 in cancerous tissues of 40 patients with NSCLC was significantly lower than that in normal lung tissues(?~2=8.571,P=0.003).The expression of TRIM35 in cancerous tissues was not significantly different from that in normal tissues or para-cancerous(?~2=2.650,P=0.104;?~2=1.867,P=0.721)(see Table 3).The clinical parameters of 148 patients with non-small cell lung cancer(see Table 1)were analyzed.The result of statistical analysis show that the expression level of TRIM35 was correlated with lymph node metastasis,and the expression of TRIM35 in patients with lymph node metastasis was significantly lower than that in patients without lymph node metastasis(?~2=13.865,P<0.01).The expression of TRIM35 in patients with late TNM stage was significantly lower than that in patients with early TNM stage(?~2=9.703,P<0.01).However,there was no significant correlation between TRIM35 expression level and differentiation,age,sex,pathological type,smoking history,drinking history and family history(see Table 2).The survival rate and recurrence rate of 148patients with NSCLC were analyzed by single factor analysis.The results showed that the overall survival rate of high expression group of TRIM35 was significantly higher than that of low expression group(?~2=93.261,P<0.01).The recurrence rate of low expression group was significantly higher than that of high expression group(?~2=73.890,P<0.01).Cox multivariate regression model was used to analyze the statistical variables in univariate analysis.The results showed that the low expression of TRIM35 was an independent risk factor affecting the survival time of patients with non-small cell lung cancer(HR=0.126,95%CI confidence interval:0.077-0.208,P<0.01)(see table 4).At the same time,the low expression of TRIM35 is also a high risky factor for relapse of NSCLC(HR=0.071,95%CI confidence interval:0.032-0.157,P<0.01)(see Table 5).Moreover,Cox multivariate regression analysis showed that lymph node metastasis was also a risk factor for recurrence after surgery.Conclusion:1.TRIM35 was expressed in non-small cell lung cancer(NSCLC)tissues,adjacent tissues and normal tissues.However,it is noteworthy that the expression of TRIM35 in cancer tissues was significantly lower than that in corresponding normal tissues,while there was no significant difference between para-cancerous tissues and corresponding normal tissues.The expression of TRIM35 was correlated with TNM stage and lymph node metastasis.This suggests that the high expression of TRIM35 may inhibit the invasion and metastasis of tumor cells.2.In patients with non-small cell lung cancer,the overall survival rate was higher and the recurrence rate was lower in the high expression group of TRIM35 than low expression.These results suggest that the low expression of TRIM35 is an independent risk factor for survival and recurrence of NSCLC patients after operation,which suggests that TRIM35 may become a new target of gene therapy.and may be a useful prognostic indicator for patients with non-small cell lung cancer.
