Objective: To investigate the effects of cinnamaldehyde on the expression of Caspase-3 and XIAP in host cells infected with respiratory syncytial virus, and provide a theoretical basis for the treatment of respiratory syncytial virus with clinical application of cinnamaldehyde. Methods: Hela cells were divided into 5 groups:(1)the normal cells group(2)RSV-infected cells were virus group(3)the cells infected with RSV were treated with low, medium and high concentrations of cinnamaldehyde as experimental group. The expression of Caspase-3 protein and XIAP protein was detected by Western blotting in the following four cases(direct inactivation stage, biosynthesis stage, virus adsorption stage and drug pretreatment)with cinnamaldehyde.Results:1.Western blot analysis of Caspase-3 protein expression, there are differences between the five groups.Compared with the normal cell control group, the expression of Caspase-3 protein in the virus group was on the rise.Compared with the virus group(except for the drug pretreatment mode),the experimental group showed a decrease in the expression of Caspase-3 protein, and the high concentration group decreased the most, followed by the middle concentration, and the low concentration decreased the least, which was statistically significant(P < 0.05).2.The expression of XIAP protein was determined by Western blot. The expression levels of XIAP were different among the five groups.Compared with the normal cell control group, the XIAP protein expression of the virus group showed a downward trend.Compared with the virus group(except for the drug pretreatment mode),the experimental group increased the expression of XIAP protein, and the high concentration group increased the most, followed by the middle concentration, and the low concentration increased the least, which was statistically significant(P< 0.05).Conclusions:1.From the change of the expression levels of proapoptotic protein Caspase-3 and anti-apoptotic protein XIAP,cinnamaldehyde has anti-respiratory syncytial virus effect.2.Cinnamaldehyde has a therapeutic effect on host cells infected with respiratory syncytial virus, and its mechanism may be related to inhibition of Caspase-3 protein expression and promotion of XIAP protein expression.
