Purposes:To detect the positive expression rate and expression of Hiwi protein in ovarian cancer tissues and normal ovarian tissues,and to analyze the correlation between Hiwi protein and ovarian cancer patients' s clinicopathological features and to explore the relationship between Hiwi protein and ovarian cancer.To detect the expression level of Hiwi mRNA in ovarian cancer tissues,and further verify the possible role of Hiwi protein in the occurrence and development of ovarian cancer at the molecular level.Follow-up of the survival rate of patients with ovarian cancer,and explore the role of Hiwi protein in evaluating the prognosis of patients.Method:A total of 49 cases of ovarian cancer tissues from January 2010 to 2012 in the People's Liberation Army 401 Hospital and the Affiliated Hospital of Qingdao University were collected(29 cases of serous carcinoma,16 cases of mucinous,4 cases of other types;19 cases of stage I to II,III ~ 30 cases in stage IV)to do SP staining.All patients underwent cytoreductive surgery without any treatment before surgery,and the control group had 10 normal ovarian tissues.1.What was used to detect the positive expression rate of Hiwi protein is immunohistochemical SP method in ovarian cancer tissues and normal ovarian tissues.2.To analyze the correlation of ovarian cancer patients between clinicopathological features and Hiwi protein expression,especially the relationship with age,clinical stage,tissue type and degree of differentiation.3.QRT-PCR method was used to detect the relative expression of Hiwi mRNA in ovarian cancer tissues.The role of Hiwi gene in the development and progression of ovarian cancer was analyzed at molecular level and its possible molecular mechanism.4.To follow-up the survival time of patients with ovarian cancer,calculate the survival rate of patients,plot the survival curve of patients,and analyze the role and clinical significance of Hiwi protein in predicting the prognosis of patients.Result : 1.Immunohistochemistry results showed that Hiwi protein was mainly distributed in cancer cell membrane and cytoplasm,no nuclear staining,and staining with different intensity of brownish yellow or brown.Among the 49 patients with ovarian cancer,37 were positive for Hiwi protein expression and 12 were negative for expression.The positive expression rate of Hiwi protein was 75.5% in ovarian cancer tissues,while the positive expression rate in normal ovarian tissues was 35%.The difference has statistical significance(P <0.05).2.The positive expression of Hiwi protein in ovarian cancer tissues was related to clinical stage,differentiation,lymph node metastasis and omental metastasis(P<0.05),but not related to patient age and tissue type(P>0.05).3.Real-time PCR results showed that the relative expression of Hiwi mRNA in ovarian cancer tissues was significantly higher than that in normal ovarian tissues.4.49 patients with ovarian cancer were followed up.Of the 37 patients with Hiwi protein positive,23 died,the mortality rate was 62.16%,and 12 of the 12 patients with Hiwi protein negative died.The mortality rate was 33.33%.Log-rank test results showed that Hiwi high protein expression was negatively correlated with postoperative survival rate(Log-rank = 3.90,P = 0.048).Conclusion : Hiwi protein is highly expressed in ovarian cancer tissues,and is related to clinical stage,differentiation,lymph node metastasis,and omental metastasis,but not related to patient age and tissue type.The high expression of Hiwi protein is negatively correlated with the survival rate of patients with ovarian cancer,and can be used as an important index to evaluate the prognosis of patients with ovarian cancer.High expression of Hiwi protein in ovarian cancer tissues provides new ideas for the study of ovarian cancer pathogenesis,clinical treatment and prognosis prediction..
