Objective:1.To detect the expression of stem cells and radiotherapy resistance-related proteins in nasopharyngeal carcinoma tissue chips.2.To explore the correlation of the three proteins in nasopharyngeal carcinoma tissue stem cells by tseting the expression of PTEN,?-catenin and Nanog.3.To discuss the correlation between PTEN/?-catenin/Nanog signal pathway and the prognosis of radiotherapy for nasopharyngeal carcinoma.Methods:Preparing the nasopharyngeal carcinoma tissue array,immunohistochemistry was used to examine the differential expression of PTEN,?-catenin and Nanog on a tumor issue microarray and X~2 test was used to analyze the correlation between two proteins.Kaplan-Meier method was used to analyze the relationship between the expression of three proteins of PTEN/?-catenin/Nanog and the survival period of patients with nasopharyngeal carcinoma after radiotherapy.Results:1.Expression of PTEN,?-catenin and Nanog:immunohistochemistry showed that negative immunostaining of PTEN was found in 62 of 130 tumors(48%).Thirty-eight of sixty-two(61%)specimens with negative PTEN staining displayed both nucleus?-catenin and Nanog immunoreactivity.2.The nuclear expression of?-catenin in nasopharyngeal carcinoma was positively correlated with the nuclear expression of Nanog(P<0.05).The negative expression ofPTEN was negatively correlated with the nuclear expression of?-cateninand Nanog(P<0.05).3.Nasopharyngeal carcinoma patients with PTEN+/?-catenin(nuclear)-/Nanog-have a long survival after radiotherapy,the effect of radiotherapy is better.While the nasopharyngeal carcinoma patients with PTEN-/?-catenin(nuclear)+/Nanog+have shorter survival period and have a poor prognosis after radiotherapy.Conclusion:1.The expression of PTEN was negative in patients with radiotherapy resistance in nasopharyngeal carcinoma.The transfer of?-catenin from the cytoplasm into the nucleus can activate the expression of the stem cell transcription factor Nanog,thereby maintaining the self-renewal of the stem cells.2.Nasopharyngeal carcinoma patients with PTEN-/?-catenin(nucleus)+/Nanog+have a high resistance to radiotherapy.The expression of this pathway can be used as an important indicator of prognosis in patients with nasopharyngeal carcinoma.
