Objective:The expression levels of NLRP3,NLRP6,NLRP12 receptor and downstream factor ASC in colonic tissues of patients with ulcerative colitis were detected.To explore the correlation between the above factors and ulcerative colitis.Methods:Thirty-one patients with ulcerative colitis were collected,and paraffin-embedded specimens of colon tissue were collected after colonoscopy.Thirty paraffin-embedded specimens of normal tissues adjacent to colon cancer were collected.The expression levels of NLRP3,NLRP6,NLRP12 and ASC between colon tissue in patients with ulcerative colitis and normal tissues adjacent to colon cancer by immunohistochemical staining and QPCR.Finally,statistical methods were used to compare the differences between the two groups.Results:The results of immunohistochemical staining:compared with the control group,the average integral optical density of NLRP3 and ASC in ulcerative colitis group was higher,while the average integral optical density of NLRP6 was lower,and there was significant difference between the two groups(P < 0.05).The average integral optical density of NLRP12 showed no significant difference between ulcerative colitis group and control group(P > 0.05).The results of QPCR:compared with the control group,the mRNA expression levels of NLRP3 and ASC were up-regulated in the ulcerative colitis group,and the mRNA expression level of NLRP6 was down-regulated,the difference was statistically significant(P<0.05).The mRNA expression level of NLRP12 was not significantly different between the ulcerative colitis group and the control group(P>0.05).Conclusion:The expression level of NLRP3 and ASC in colon tissue of patients with ulcerative colitis was increased,the expression level of NLRP6 was down-regulated,and the expression level of NLRP12 was not significantly different.The high expression of NLRP3 and ASC may be one of the pathogenic factors of ulcerative colitis.NLRP6 may play a protective role in the occurrence and development of ulcerative colitis.
