As we all know,cancer has become one of the diseases that threaten human health,ranking second only to cardiovascular disease.We know that traditional cancer treatment methods have strong side effects on the human body.Finding reasonable methods and high-efficiency and low-toxic drugs to prevent and treat cancer is a hot topic in medical experts at home and abroad,and it is also a mainstream topic in the pharmaceutical industry.The world's marine resources are extremely rich,and species diversity provides a material basis for the study of new anti-tumor drugs.In recent decades,a variety of anti-tumor active substances have been discovered and used in the clinic.Among them,Meretrix meretrix Linnaeus has been used to treat cancer for a long time.This paper introduces the anti-tumor effect of Meretrix meretrix Linnaeus on the basis of predecessors.We isolated a new active substance from its body fluid and studied its antitumor activity.The research team adopted a series of biological separation techniques,including ammonium sulfate fractionation,ion exchange chromatography,hydrophobic chromatography,gel chromatography,etc.,which separated and purified the body fluid of Meretrix meretrix Linnaeus and obtained a new active substance,hasing a good inhibitory effect on tumors.The molecular weight of the polypeptide was 15 KDa and identified as MM-15 by SDS-PAGE gel electrophoresis.The BCA method was used to detect the protein concentration,and using MTT assay to detect the growth inhibition activity of MM-15 on tumor cell lines?human lung cancer A549,human liver cancer BEL-7402,human colorectal cancer HCT-116,human ovarian cancer HeLa cells,human pancreatic cancer Aspc-1?and normal cell lines?breast cell MCF-10A and mouse embryonic fibroblast NIH-3T3?.In the above order,the IC50 value of MM-15 to the above cell line was 31.80?g/mL,47.75?g/mL,64.3?g/mL,46.00?g/mL,45.13?g/mL,149.51?g/mL,and 123.13?g/mL.The results showed that MM-15 has broad-spectrum anti-tumor activity,and the inhibitory effect of MM-15 on human lung cancer A549 is the best,but it has no obvious effect on two kinds of normal cells.Subsequently,human lung cancer cell A549 was taken as the research object to explore the effect of MM-15 concentration and action time on the proliferation of cancer cells.The results showed that the inhibition of A549 cells increased with the increase of MM-15 concentration,the IC50 was 30.30?g/mL;the effect of MM-15 was the best after two days of A549,and the efficacy lasted for more than three days.The study on the anti-tumor activity of Meretrix meretrix polypeptide MM-15lays a material foundation for its anti-tumor mechanism research and gene recombination,and opens up a new way for the treatment of malignant tumors,which has broad prospects for development.
