Abstract/Summary:
Objective: Patrina scabiosaefolia Fisch.ex Trev.,a plant of Valerianaceae,is the dried whole plant of Patrina scabiosaefolia Fisch.ex Trev.As a a traditional Chinese medicine,it has the functions of clearing away heat and detoxification,eliminating swelling and abscess,activating blood circulation and removing blood stasis.Through screening the extract layer of Patrina scabiosaefolia,which has the activity of promoting intestinal contraction,separating the monomer compounds,and exploring the mechanism of Patrina scabiosaefolia which can promote intestinal contraction,which will provide a certain experimental basis for the development of Patrina scabiosaefolia monomer compounds into preparations and clinical medicines.Methods: In this paper,Patrina scabiosaefolia extract was taken as the research object.According to the previous experimental conclusion.Patrina scabiosaefolia extract was carried out under the optimum extraction conditions.The effects of total extracts on the contraction of duodenum,jejunum,ileum and colon in SD rats were studied by means of in vitro intestinal experiment.Then choosing the best polar extracts for promoting intestinal contraction by examinating the acceleration rate of each polar extract to intestinal contraction.Separating the active fracion by column chromatography and PHPLC then identified the chemical constituents in the active fraction.Results: The effects of total extract of Patrina scabiosaefolia on the contraction of duodenum,jejunum,ileum and colon in SD rats were studied by in vitro intestinal experiment.The results showed that Patrina scabiosaefolia total extract had the most significant effect on promoting the contraction of colonic smooth muscle.On this basis,combined with the yield of polar extracts,the effects of four different polar extracts of Patrina scabiosaefolia on contraction of colonic smooth muscle in SD rats were studied.The results showed that the extracts of Patrina scabiosaefolia had contractionpromoting effects on colonic smooth muscle in SD rats.The activity of different extracts was dichloromethane layer > ethyl acetate layer > water layer > n-butanol layer.Dichloromethane extract,ethyl acetate extract,n-butanol extract and water extract were 26.54±2.73 %、15.59±3.05 %、6.74±3.67 %、10.78±3.24 % respectively,which indicated that dichloromethane extract had the most significant effect on the contraction of colon.Four extracts from different polar layers of Patrina scabiosaefolia at the same concentration were investigated.The results showed that Patrina scabiosaefolia dichloromethane extract had the most significant effect on promoting the contraction of colonic smooth muscle.Based on the above experimental results,the dichloromethane extract of Patrina scabiosaefolia was selected as the active fraction to further study its composition.22 monomer compounds were isolated from the extraction layer of dichloromethane,and they were identified by physical and chemical properties and modern spectroscopy.They are oleanolic acid(1),ursolic acid(2),palmitic acid(3),dodecanoic acid(4),α-tocopherolquinone(5),maslinic acid(6),4-hydroxy-3-methoxybenzaldehyde(7),2,6-dimethoxy benzoquinone(8),3β-acetyloxy-12-en-28-ursolic acid(9),α-tocospiro B(10),22E-3β-hydroxy-5α,6α-epoxyergosta-22-en-7-one(11),2α-hydroxy ursolic acid(12),linoleic acid(13),β-sitosterol palmitate(14),3-hydroxy-1-(4′-hydroxy-3′-methoxy-phenyl)-1-propanone(15),burselignan(16),hederagenic acid(17),6-methoxy-7-(1′-henicosacyloxy-3′-hydroxy-2′-propoxy)coumarin(18),(+)-isolariciresinol(19),castanopsol(20),3β-hydroxy-11α-methoxy-olean-12-en(21),tetradecanoic acid 2,6,6-trimethyl-1-(3′-oxo-but-1′-enyl)-7-oxa-bicyclo[2.2.1]hept-2-yl ester(22).After searching the Scifinder and Reaxys databases,compound 18,22 was not published and is a new compound.Compounds 5,7,8,10,11,15,16 and 19 were isolated from the genus Patrina for the first time.Conclusion: In this paper,the intestinal peristalsis-promoting activities of different polar extracts of Patrinia scabiosaefolia Fisch.ex Trev.were studied.It was found that dichloromethane layer of Patrinia scabiosaefolia had the highest activity on promoting the contraction of colonic smooth muscle and 22 monomer compounds were isolated from it.Two of them were new compounds,and 8 of them were isolated from the genus Patrina for the first time.which laid a solid foundation for further evaluation of Patrinia scabiosaefolia in the treatment of gastrointestinal diseases.
