Objective:By observing the effects of high,medium and low doses of Jiawei Wendan Tang on the expression of serum leptin(LEP),adiponectin(ADP)and tissue AMPK/ACC/CPT1 signal pathway-related proteins and genes in nutritional obese female rats.To explore the mechanism and dose-effect relationship of this prescription in improving lipid metabolism in Nutritional Obese rats.Methods:100 SD female rats were randomly divided into 5 groups according to body weight.They were normal group,model control group and Jiawei Wendan Tang high,medium and low dose groups.Establishment of a nutritional obese rat model using the "general feed+high fat emulsion+carbonated beverage" feeding method.Re-scrambling the grouping after successful modeling,administration by gavage,while administering the drug,the high-fat emulsion is still administered,anesthetized rats after 5 weeks of continuous administration.The levels of serum LEP,ADP,AMPK,ACC and CPT1 in liver and gastriculus muscle tissues were determined by Elisa.Real-time PCR was used to detect the expressions of LEPmRNA and MCAmRNA in hypothalamic tissue,AMPKβmRNA and ACC2 mRNA in liver,AMPKα1mRNA in adipose tissue and AMPKα2mRNA in gastriculus muscle tissue in each group.Research results:Compared with the model control group,the high-dose group of Jiawei wendan Tang significantly inhibited the expression of CPT1 and AMPKα2mRNA in the gastriculus muscle tissue of rats.The medium dose group can significantly inhibited the expreesion of CPT1 in gastrocnemius muscle tissue,LEPmRNA,MCAmRNA in hypothalamic tissue,AMPKα1mRNA in adipose tissue and gastrointestinal muscle tissue AMPKα2mRNA of rats.The low-dose group significantly inhibited the expression of serum LEP,hypothalamic LEPmRNA,MCAmRNA,AMPKα1mRNA in adipose tissue,AMPKα2mRNA in gastriculus muscle tissue,and AMPKβ and ACC2 mRNA in liver tissue.Compared with the high-dose group of Jiawei wendan Tang,the expression levels of LEPmRNA,MCAmRNA in hypothalamic tissue and AMPKα1mRNA in adipose tissue in the medium dose group were significantly higher than those in the high dose group.The expression levels of LEPmRNA in hypothalamic tissue and AMPKα1mRNA in adipose tissue of the low-dose group were better than those in the high-dose group.All the above results were statistically significant(P<0.01 or P<0.05).Conclusion:The mechanism of Jiawei Wendan Tang against nutritional obesity in female rats is related to regulating the expression of AMPK in liver,gastrocnemius muscle and adipose tissue,intervention of LEP resistance,reducing the expression levels of ACC2 mRNA and MCAmRNA in liver tissue and brain tissue.The middle and low dose groups have better effects.
